A methodology of chemical kinetics was introduced to the investigation of the regulation network involved in DNA synthesis during rat liver regeneration following partial hepatectomy. Findings are as follows : 1. The rate-limiting enzymes in the DNA synthesis are thymidine kinase (TK) and the folate-dependent thymidylate synthase (TS), which catalyze the formation of thymidylate via the salvage and de novo pathways, respectively. 2. By evaluating the effect of drugs administration and the ablation of endocrine organs on activities of TS and TK which are S-phase specific enzymes, it was shown that the transition of the liver cells from G_1 state to S-phase takes place 10-12 h after partial hepatectomy, and was regulated by catecholamines released from the splanchnic nerves through hepatic α_1-receptors, and utilized calcium ion and metabolites of inositol turnover as intracellular messengers. 3. TS was purified from regenerating rat liver and characterized. Antiserum of TS was raised and used to establish the immunoblotting assay. The rise of TS activity during liver regeneration was due to the induction of TS and not to the activation by calcium ion.

Using human mustache, the excretion of intracorporeal methylmercury by the administration of thiamin tetrahydrofurfuryl disulfide (TTFD) was investigated. The usual dosage of TTFD administered orally significantly increased the mercury level in the mustache seven to ten days later, but another thiamin derivative, S-benzoylthiamin monophosphate (BTMP) did not show such effects. From their chemical structural difference it was strongly suggested that the mechanism of the mercury excretion was based on the side chain structure of TTFD, furfurylmercaptan.

Vitamin K_3 in foods derived from livestock was measured by a method based on high-performance liquid chromatography (HPLS) combined with coulometric reduction and fluorometric detection. The vitamin extracted from foods was pirified with a Sep-Pak silica cartridge and/or a Sep-Pak silica cartridge followed by thin-layer chromatography, respectively, and then measured by using an HPLC system. The detection limit of vitamin K_3 was 50 pg/g or ml in foods.

A rapid and reproducible procedure using extraction system with ethyl acetate /n-hexane(1:9) after saponification, is described for the determination of retinol in foods by the reversed phase high performance liquid chromatography. The results obtained by the proposed procedure were comparable to those by the procedure using the ethyl ether extraction system. Recoveries of retinol from egg white and pork separable fat were 99.1 and 92.2 %, respectively. The coefficients of variation were 3.0% and 0.9% for 5 replicate analyses of process cheese and infant formula, respectively.

The determination method of NADP, which has been reported previously by Shibata et al. [Agric. Biol. Chem., 50,2941-2942(1986)] was successfully improved for the estimation of blood NADP. Blood NADP levels in 40 women students were measured by using this improved method. The μl of whole blood was required for this method of NADP determination. Ten minimum blood NADP value was 6.9 nmol/ml of whole blood and the maximum value 13.3 nmol. NADP levels of about 80% student were included within 8-11 nmol/ml of whole blood. The mean ± S.D. was 9.6 ± 1.4 nmol/ml of whole blood.
