    The following phosphate derivatives of vitamine B_6 were prepared in crystalline form : pyridoxal-5-phosphate, pyridoxamine-5-phosphate, pyridoxine-5-phosphate and deoxypyridoxine-5-phosphate. Phosphorylation was accomplished by the reaction of pyridoxamine or isopropylidenepyridoxine or deoxypyridoxine with a mixture of phosphoric acid and phosphorus pentoxide. Pyridoxal-5-phosphate was obtained in 72% yield by the deaminative oxidation of pyridoxamine-5-phosphate with glyoxylic acid and copper or nickel salt which was the highest among several kinds of synthetic method.

    We studied on the ralationship between electrocardiographic changes and thiamine contents in blood and heart muscles in thiamine deficient rats. In rats in which there were eloctroardiographic changes, thiamine contents decreased extremely (from 1/3 to 1/10 of normal contens). However, below the decreased level, no relationship between electrocardiographic changes and thiamine contents was observed. The authors assume that for thiamine deficient cardiac disturbance to appear it is recessary that thiamine contents decrease below a certain level (at least 1/3 of normal contents). As there was no direct relationship between degree of cardiac symptoms and thiamine contents it is assumed that cardiac disturbance is related to the degree of metabolic disturbances owing to thiamine deficiency.

    Deficiency symptoms of B vitamins were observed under the Japanese field conditions among the meat-type chicks imported recently for the purpose of broiler production. Accordingly, the requirements of thiamine, riboflavin, pyridoxine, niacin and choline for White Rock chicks, the third generation of chicks imported from Abor Acore Farms, U.S.A., were studied and from the growth rate and deficiency symptoms it was found that the higher dietary levels of riboflavin, niacin and choline than those recommended by National Research Council, U.S.A., were required by the chicks. In the second experiment, niacin-deficient diets with an addition of graded levels of niacin were fed to the same White Rock chicks for 3 weeks. Linear relationship between body weight gain and logarithm of dietary niacin level was observed. The intersection of the regression line and y=200.3g, i. e. the average gain of chicks fed much excess of niacin, gave the estimate of niacin requirement. This was 30 mg/kg diet, or 1.1 times more than the recommended level.

    Since the previous work revealed the higher requirement of riboflavin for White Rock chicks, the requirement of the vitamin for the chicks of the same strain was studied. Two types of riboflavin-deficient basal diets, purified and practical, were supplemented by various levels of riboflavin and fed to day-old chicks for 3 weeks. Typical riboflavin-deficiency symptom, curled-toe paralysis, was observed among the chicks fed with the diets containing riboflavin of about 2.9 mg/kg diet or less. Linear relationship between body weight gain and logarithm of riboflavin level in the diets of both types was observed and the requirement of riboflavin was estimated by the same way mentioned in the previous paper for niacin, which was 6.3 mg/kg with purified diet and 4.0 mg/kg with practical type diet. Although chicks fed with purified diet took about 9% less diet than those fed with practical type diet, the difference in riboflavin requirement between the types of the diet was assumed mostly due to the experimental error among the individual chicks.

    0.5% of thiamine terahydrofurfuryldisulfide (TTFD) is applied to the mucosa of jejunum of anesthetized dogs from which the metenseric nerves has been cut, an intra-mucousal reflex occurs, and the peristalsis of the oral side to the point of application increases while that of the caudal side is suppressed. When 1.5mg to 5 mg/kg of TTFD is injected intravenously, a marked increase in the peristalsis of the small bowel occurs 2 to 3.5 minutes later. Since there is a relatively long time lapse before this stimulative effect is seen, it is felt that this effect is due to an intermediate compound which is formed when TTFD is changed to thiamine.

    Some analogs of toxopyrimidine with ethyl group attached to 2 position of the pyrimidine ring, were prepared. The synthesis of 2-ethyl-4-amino-5-hydroxymethylpyrimidine (III) started with the condensation of propioamidine with 2-methoxyethoxymethyl-3-ethoxypropionitrile to yield 2-ethyl-4-amino-5-propioamidomethylpyrimidine (I), which gave (III) through 2-ethyl-4-amino-5-aminomethylpyrimidine (II). Condensing propioamidine with 2-methoxymethylene-3-ethoxypropinitrile afforded 2-ethyl-4-amino-5-ethoxymethylpyrimidine (IV) which yielded 2-ethyl-4-amino-5-bromomethylpyrimidine (V) on treatment with hydrobromic acid. Catalytic hydrogenolysis of (V) gave the 5 methyl-derivative (VI). 2,5-Diethyl-4-amino-6-hydroxypyrimidine (VII) was prepared by condensing propioamidine with α-cyanobutyrate. Upon chlorination with phosphorus oxychloride and phosphorus pentachloride, (VII) gave the 6-chloro-derivative (VIII), which was treated to give 2,5-diethyl-4-aminopyrimidine by catalytic hydrogenolysis. Biological action of these compounds on animals or microbes will be reported elsewhere.

    As preparatory studies of the kinetics on carboxylase catalysis, partial purification of apocarboxylase and some properties of the enzymatic reaction were investigated. Apocarboxylase solution obtained from bakers' yeast by repeated extractions with 0.05M Na-phosphate buffer was purified to approximately 120-fold by gel-filtration with Sephadex G-25 and by column-chromatographic separation with DEAE-cellulose. The purified enzyme was tinged with pale yellow, and showed the specific absorption-bands at 405mμ and 278mμ. The enzymatic reaction took place only when the two cofactors, cocarboxylase and Mg^<2+>, was added in the reaction mixture. Mg^<2+> and cocarboxylase combined with apocarboxylase directly, and they were increased the affinities for apoenzyme by each other. The enzymatic reaction was remarkablly affected by ionic strength of medium. So that it was observed that the inhibitory effect of ions was caused by substrate.

    In the course of study on the biological effect of AICA, it was recognized that the pyruvate decarboxylation by yeast apocarboxylase and cocarboxylase system was accelerated by addition of AICA to the reaction system. This effect was not observed in the yeast carboxylase extracted without cocarboxylase elimination. It was shown that this effect was different from the mechanism of Ochoa's effect based on thiamine addition and of the prevention of heavy metal inhibition. The effect of AICA is supposed to be concerned in the formation of apo- and coenzyme complex.

    This paper deals with studies on natural forms of vitamin B_<12> active substances in the cells of thermophylic methane bacteria. A purified extract was obtained by an extraction with hot alcohol and subsequent phenol treatment, instead of the usual extraction method with cyanide solution in order to avoid the transformation of cyanide-labile vitamin B_<12> analogues. The extract exhibited the coenzyme actvity in Abeles・Lee's intramolecular oxidation-reduction system. By paper ionophoresis performed in 0.5N acetic acid, five active zones on E.coli vitamin B_<12>-less mutant were observed ; three of them (tentatively named P1-P3 in order of mobolities) moved toward the cathode, one (P4) stayed near the origin, and the fifth (P5) migrated toward the anode. By cyanide treatment, disappearances of P1 and P2 and simultaneous increase in area of P4 were observed. In bioautogram using Ochromonas malhamensis, growth zones corresponding to P2 and P3 were not detected. From these results, it seems appropriate to conclude as follows : P1 is DBCC ; P2,non-cyano type B_<12> analogue lacking 5,6-dimethylbenzimidazolyl cobamide structure ; P3,pseudocobalamin ; P4,cyanocobalamin and P5,an unidentified protein-bound form with anionic character.

    This paper describes the formation of DBCC and other unidentified vitamin B_<12> analogues from cyanocobalamin during thermophilic anaerobic digestion of waste distillage of acetone-butanol fermentation factory. After 24 hours of addition of cyanocobalamin, mixture of bacterial cells and sludges was centrifuged and extracted with hot ethanol. The alcoholic extract was concentrated in vacuo, purified by phenol treatment and passed through DEAE-cellulose column. Elution was carried out successively with water (orange-red colored fraction, tentatively named DC-l), 0.1 N acetate buffer (pH 4.7) (orange-red colored, DC-2), and 0.1 M NaCl・acetone mixture (brown colored, DC-3). DC-1 showed vitamin B_<12> activity on E.coli No. 215 and coenzyme activity in Abeles・Lee's intramolecular oxidation-reduction system. From the absorption spectrum and its changes by light and cyanide, and the behaviors in paperionophoresis, the identity of DC-1 with DBCC was concluded. The yield of DBCC was about 20% of added cyanocobalamin. DC-2 and DC-3 exhibited the growth effect on E.coli, though lower than that of DBCC, but no coenzyme activity in Abeles・Lees system. From the results of chromatography, spectroscopy and paperionophoresis, it was considered that DC-2 consisted of two unidentified cobamides and DC-3 contained unidentified analogues or decomposition products of cyanocobalamin.
