Some properties of folic acid γ-glutamyl hydrolasc (conjugase) [EC 3.4.22.12] and dihydrofolate reductase [EC 1.5.1.3] in silkworm larval bodies were investigated. The conjugase enzyme had an optimum pH of 7.8, and an optimum temperature of 50℃. The enzymatic activity was stimulated in the presence of 2-mercaptoethanol, K^+ and Mg^<2+>, but inhibited by p-hydroxymercuriphenyl sulfonate, Co^<2+>, Zn^<2+> and Fe^<2+>. This conjugase converted pteroylpenta-γ-L-glutamic acid to pteroyl monoglutamic acid exclusively, and the Km value was about 9.6mM when measured at pH7.8, 37℃ with pteroyltriγ-L-glutamic acid as the substrate. The dihydrofolate reductase in the silkworm fat body required NADPH, had an optimum pH of 4.5〜7.5, and an optimum temperature of 35℃. Its actvity was stimulated in the presence of 2-mercaptoethanol, guanidine, Mg^<2+> and high concentration of K^+ and Na^+, but inhibited by p-chloromercuribenzoate, formamide and Co^<2+>. The Km value measured at pH5.0, 30℃, was about 2.7mM when dihydropteroyl glutamic acid was used as the substrate.

In urine the vitamin C values obtained by using the α,α'-dipyridyl method were often elevated due to urinary reducing substances. Ascorbate oxidase is highly specific for oxidizing vitamin C. An improved method using ascotbate oxidase is described for the determination of urinary vitamin C. The method is specific, sensitive and reproducible, and it is also rapid and simple enough for many measurements. Using the method the urinary vitamin C was determined for 176 male students, 61 female students, 352 male staff members and 81 female staff members of our university. There was little difference in overall frequency distribution of vitamin C values in the four groups. However, somewhat lower vitamin C levels were found in students compared with staff members, and in male groups compared with female groups.

Spinach nutrient levels seem to change considerably throughout the year. In this paper, we measured the levels of proximate composition, minerals, carotene, niacin and vitamin C in spinach in spring, summer and winter. The water content in summer was significantly high (p<0.001) compared to that in other seasons, while the levels of protein and carbohydrates were low (p<0.001). Levels of lipid and ash tended to increase in spring. Mineral contents depended on the individual specificity. Vitamin content decreased in summer. In particular, the levels of niacin and vitamin C were significantly lower in summer.
