Examining the nutritional requirements for B.thiaminolyticus and its related organisms, thiamine was found to be essential for B.thiaminolyticus, B.alvei, B. larvae and B.circulans, further, it was demonstrated that cysteine or cystine promoted the growth of these organisms.

In order to study the mechanism of the oxidation of ascorbic acid by metal ion, ascorbic acid solution was oxidized with cupric or ferrous ion by the aeration at room temperature. The reacting solution became brown, and oxalic acid, L-threonic acid and carbon dioxide were detected as oxidized products and the existence of some reductones was supposed. From above results, the decomposition mechanism of ascorbic acid by metal ion was supposed as indicated in Figure 2.

Chemical changes of ascorbic acid in the procedures of black tea manufacture were investigated. Ascorbic acid in tea leaves was oxidized and decreased in amount in all stages of manufacture and two thirds of ascorbic acid were oxidized beyond 2,3-diketogulonic acid. L-Threonic acid, glyceric acid, glycolic acid and oxalic acid were detected as oxidized products of ascorbic acid from the fermented tea leaves, to which much ascorbic acid was added. It was therefore, supposed that a part of ascorbic acid in tea leaves was oxidized mainly by polyphenol oxidizing enzymes during the black tea fermentation, followig the pathway of the decomposition of ascorbic acid which was studied and reported by these authors.

The improved thiochrome method using benzenesulfonyl chloride as thiamine inactivating agent was proposed by O.Bessey (J.Biol.Chem.195,453,1952). We checked this method and one drop of benzenesulfonyl chloride was proved sufficient to decompose 50 μg thiamine completely in alkaline solution. A sample solution (I) containing thiamine can be converted into thiochrome by alkaline ferricyanide solution, while another solution (II) containing the same amount of thiamine, after alkaline benzenesulfonyl chloride treatment does not produce thiochrome on ferricyanideoxidation. Excess of ferricyanide is both decolorized by alcoholic H_2O_2 solution, then a thiochrome standard solution (0.1 mg% in 0.01N HCl) is poured into the latter solution (II) drop by drop until its fluorescence becomes the same strength as the former (I) by comparing them under ultraviolet ray. Thus thiamine can be estimated by the simplified method using thiochrome water solution without isobutanal extraction. Crystals of mp. 113-114℃ were isolated as the reaction product of thiamine and benzenesulfonyl chloride, as we published it this journal 13.

Formerly, one of the authors found that the indophenol-butanol solution was very stable and applying this stability, he proposed an indophenol-butanol titrimetric method for determination of vitamin C. The drawback of the former indophenol method due to the instability of the indophenol aqueous solution, was thus obviated. Recently, the authors confirmed spectrophotometrically the stability of indophenol-butanol solutions, that is to say, even such a thin solution as that of 3mg% was stable for 8 weeks and moreover it was not discolored by acids. Applying the stability of the above solution to the colorimetric method, the authors established the indorphenol-butanol colorimetric method. Moreover the authors confirmed that, in this method, p-chloromercuribenzoate, which was proposed by Owen et al. as removing reagent for SH-compounds, completely removed not only such SH-compounds as reduced glutathione and cysteine but also H_2S, Na_2S_2O_3 and Na_2SO_3. By this method, the vitamin C in plant tissues and human blood could be determined very accurately, and values obtained by this method agreed well with the measured by the ascorbic acid oxidase method.

A survey of carotenogenic activity of 72 fungus strains were conducted. In addition to the strains known as carotene producers, Penicillium sclerotiorum, P.multicolor, P.lapidosum, and Rhisoctonia sp. Y-08 were found to produce carotene when grown on Difco wort agar media. P.sclerotiorum NRRL 2074 produced about 0.7 mg total carotene per gram of dry mycelium and its major pigments were β- and γ-carotene.

1) The growth of Penicillium sclerotiorum NRRL 2074 reached its maximum in 10 days while the maximum carotene formation was attained in 15 days. 2) Light had a marked stimulative effect on the carotene production. 3) Shaking during the incubation period has a deleterious effect on the carotene formation. 4) Czapek-Dox agar was good for the growth but was inferior to wort agar in the carotene formation. 5) P.sclerotiorum NRRL 2074 produced about 1 mg of total carotene per gram of mycelium dry weight, of which approximately 65% was β-carotene and approximately 25% was γ-carotene. In addition, as minor pigments, α-carotene, 3-cis-isomers of β-carotene, 2-cis-isomers of γ-carotene, and zeacarotene were identified.

The children subjected to this investigation were divided into 2 groups (I and II). Group I included 8 healthy boys, and group II included 20 boys with nutritional dystrophy. Ten mg of pteroylglutamic acid was given subcutaneously and the pteroylglutamic acid excreted in the 24 hour urine after the injection, was estimated fluorometrically. The average (±fiducial limits with a level of significance of 0.5%) of the pteroylglutamic acid excreted in the urine after the injection were 6.7±1.1 mg and 4.4±0.6 mg in groups I and II respectively. The macrocytic anemia was found in 17 cases of group II and in one of group I. Both the high incidence in macrocytic anemia and the small urinary output of pteroylglutamic acid after its injection suggested that the pteroylglutamic acid deficiency existed in the children with nutritional dystrophy.

The distribution of riboflavin and other fluorescent substances in the skin was studied with 11 species of animals by paper partition chromatography. (1) The forms of riboflavin in the skin of Mammalia or Aves are considerably different from those in the skin of Amphibia. The majority of riboflavin is ester forms of the vitamin (FAD and FMN) in the former, while almost all is free form in the latter. (2) It is interesting that a blue-fluorescent substance having Rf 0.45,which was confirmed to be kynurenine by several tests, is observed in general in the mammalian skin.

Brain glutamic acid decarboxylase activity during the running fit induced by injection of a pyrimidine derivative, 2,5-dimethyl-4-aminopyrimidine, and carbonyl reagents (hydroxylamine, semicarbazide, thiosemicarbazide, isonicotinic hydrazide, benzoic hydrazide) in the white rat was compared to that found during convulsions induced by other convulsive agents (strychnine, cardiazol, ammonium chloride). The brain glutamic acid decarboxylase was reduced in the running fit induced animals and moreover, this inhibition was already present prior to the convulsion. It is suggested that inhibition of this enzyme is characteristic of the running fit and that the nucleus lenticularis and nucleus caudatus of the brain stem are the sites of origin of the running fit.

The effect of N-carbobenzoxy-L-glutamylcholine on hypertension, experimentally induced in the rabbit by a cholesterol diet, was studied from the standpoint of hypotensive effect and histopathologically. An effect was noted both prophylactically and therapeutically in the blood pressure experiments. Histologically, there was an inhibition of atheroma development in the aorta and the deposition of fat in the various organs was less in the group given the compound prophylactically. It is suggested that there was a beneficial effect on fat metabolism.
