  • シオミズツボワムシの濾水率について

    In the present study, the filtration rate and the ingestion rate of the rotifer, Brachionus plicatilis, were investigated under various culture conditions.
    Several groups of the rotifer varying in density were cultured for 60min, in an experimental medium containing a species of marine Chlorella at a definite density. The filtration and ingestion rates of the rotifer were then estimated by measuring the optical density of Chlorella as the phytoplankton density in the media. The rotifer used in these experiments was pre-cultured in two ways. In one case the rotifer was made to have food in its stomach by feeding in the experimental medium (satiated condition) and in the other case the rotifer was made to have an empty stomach (empty condition). Results obtained are as follows.
    1. When the rotifer is fed with Chlorella in the test medium kept at a density of less than about 213×104 cells/ml, the rotifer maintained its filtration rate at the highest and almost constant level of about (7-10)×10-5 (empty condition) and (5-7)×10-5ml (satiated condition) per min. per individual while the ingestion rate continued to increase. Above this density of Chlorella, the filtration rate decreased with the increase of the density of Chlorella in the test medium while the ingestion rate maintained an almost constant level of about 200 cells/min/individual under the empty condition. This tendency of ingestion rate could not be observed distinctly in the rotifer under the satiated condition.
    2. The filtration rate measured under the satiated condition showed its highest in the media having a temperature over 22.0°C, chlorinity at about 7.8% and pH value at 8.0.
    3. Assuming that the filtration rate measured at a definite Colorella density indicates the physiological activity of the rotifer under various culture conditions, the above-mentioned conditions are the most suitable to exhibit its physiological activity to the highest degree. Therefore, the above conditions seem to be the most favorable for the culture of this rotifer.

    It is difficult to calculate theoretically the tension and the configuration of the towing ropes when such exhibit three dimensional motion. In this paper, a method of approximation is described to calculate the three dimensional problem, such as the tension at the warp of a two-boat trawl or the distance between the pair of otter boards of a trawl gear, using the loop functions of the basic catenary γ', σ', ξ', η'. These are given by the formulas
    γ'=|cosecθ|=η'+1, σ'=|cotθ|=sinhξ', ±ξ'=ln|cotθ/2|, η'=|cosecθ|-1=coshξ'-1
    where θ is the inclination of the tangent of the basic catenary.

    The approximate calculation process of the three dimensional problem described in part III is too complex to be of any practical use. In this paper, the author has simplified this calculation process by using the ABC Table which was derived from the loop functions of the basic catenary σ', ξ', η' as shown by
    ±A=±σ', B=√(ξ')2+(η')2, ±C=cos-1(η'/B)
    where σ', ξ' and η' are given by
    σ'=cotθ, ±ξ'=ln|cotθ/2|, η'=|cosecθ|-1.
    By this method these approximated values agreed well with the previous experimental values of the mid-water trawlers and the stern trawlers.

    In a previous paper the seasonal change in the general vertical distribution pattern of the meduea of Aurelia aurita (LINNÈ) was described, but a more detailed qualitative as well as quantitative study dealing with its diel change has hardly been brought forth to present. The present paper, nevertheless, attempts to form a record on the above-mentioned subject.
    A series of observations were made at five stations near the mouth of the western coast of the Urazoko Bay, a small inlet on Wakasa Bay situated along the central part of Japan Sea. The observations were conducted at 13:00 to 18:00 hrs. October 17 th 1968. At about one-hour intervals, vertical distribution of Aurelia medusae was checked qualitatively with an underwater-television (Sensuiken Type No. 500), and the number swimming at each depth was recorded quantitatively by means of vertical hauls of a closing plankton-net (51cm mouth aparture, 0.33mm in mesh size). Simultaneously, meteorological and hydrographic observations were made. The results obtained are summarized as follows:
    Medusae were mainly distributed at the upper 3m layer during daytime (13:00 to 16:00 hrs.) when surface illumination was more than 103 lx, and they were distinctly abundant at the surface just before sunset. At sunset (17:00 hrs.) however, some of the medusae were observed to sink when the light intensity was less than 103 lx. After that they all showed a more conspicuous tendency to sink, and by 18:00 hrs., when it was nearly quite dark, the medusae appeared scattered from the surface to the middle and as far as the bottom layer (15 to 24m). Therefore, it may safely be concluded that underwater illimination plays an important role in the vertical distribution of the medusae and that Aurelia medusa is thus one of the twilight migrants in coastal waters.
    On the other hand, no correlation was observed between the vertical distribution of the medusae and other environmental factors such as wind direction, wave strength, current, water velocity and water temperature.

    In the season of Microcystis bloom in Lake Suwa, both the horizontal and vertical distributions of dissolved oxygen around a floating net (9×9×1.5m) with about 7000 carps were studied. Oxygen concentration in the upper waters at the edge of the net or inside it was always lower than that at the control stations at a distance of 100 or 200m from the net, and the presence of nets caused the decrease of oxygen concentration up to the distance of about 50m from the net. The decrease in oxygen concentration was more prominent in the layer of 0 to 2m (32% saturation on Sept. 17), especially when the net was set in the windward direction. Oxygen consumption of bottom deposits around the net, up to a distance of about 20m, was much larger than that at the other stations located far away.

    The process of oocyte maturation was observed in the chum-salmons, Oncorhynchus keta, captured at Otsuchi Bay and Otsuchi River, Iwate Prefecture, Japan, during the months of November 1969 and 1970. When the female salmons arrived at Otsuchi Bay, the oocytes conatined in their ovaries were either in the end of the growth period of oogenesis or in the beginning of the maturation period. From the time of arrival in the bay until the fish entered the river, a series of changes proceeded in the oocytes. The coalescence of yolk globules was first observed, followed by the migration and segregation of oil drops, the development of cortical cytoplasm, the formation of the blastodisc, the thickening of the chorion around the micropyle, the disappearence of the germinal vesicle, the meiotic division of the nucleus, and the rupture and shrinkage of the follicular envelopes. From the observation on oocyte structure, the maturation period of the oocytes can be devided into four stages, A, B, C and R. Because fish possessing fully ripe eggs in the body cavity were captured in the bay, it is ascertained that the oocytes can complete their maturation process within the bay prior to ascending the river.

    A member of the imperfect fungi was isolated on a soytone-dextrose agar medium with chloramphenicol and cycloheximide from gill lesions of Penaeus japonicus BATE infected with black gill disease. Intramusculaly inoculation of conidia of the fungus caused invariably black gill disease in prawns, and the fungus could be isolated from gill lesions of artificially-infected prawns. The gills of healthy prawns were alway free of the fungus. Therefore the fungus was regarded as the pathogenic agent of black gill disease. The fungus produces mainly 4-celled, canoe-shaped macroconidia and 1-celled microconidia. Usually the mycelium is hyaline, but it characteristically produces a brown pigment when grown on the above-mentioned medium, salt Sabouraud's medium, and a salt potato-dextrose agar medium. On the basis of morphological criteria, this fungus fits best in the genus Fusarium.

    Previously, the authors reported the sterol constituents of six species of starfishes (Asterias amurensis, Asterina pectinifera, Luidia quinaria, Asteropecten scoparius, Asteropecten polyacanthus, and Coscinasterias acutispina). ?? 7-Cholestenol was the major sterol (about fifty percent of total sterols) in these animals. ?? 7.22-Ergostadienol, ?? 7.22-stigmastadienol, and ?? 7-stigmastenol were also identified by comparing the retention times of GLC with those of authentic samples, and the occurrence of C28 sterol, stellastenol, was recognized in all starfishes investigated.
    In the present study, two unidentified sterols reported in the previous paper were examined by GC-MS spectrometry. These were proved to be cholestanol and ?? 7.22-cholestadienol, respectively. Besides, the presence of a new sterol, which was suggested to be 24-nor- ?? 7.22-cholestadienol by mass spectrum analysis, was recognized in all six species of starfishes.

    The triglycerides and cholesteryl esters of six fish oils were analysed by gas chromatography after preliminary separation by thin layer chromatography. Generally, fish oils contained C-even triglycerides from C-30 to C-60 with small amounts of C-odd triglycerides. The oils of fresh-water fish contained mainly C-even triglycerides from C-40 to C-60, while oils from salt-water fish contained in addition those from C-30 to C-40. A characteristic of salt-water fish oils was the predominance of C-odd triglycerides over C-even below C-45, in contrast to the normal predominance of C-even triglycerides above this length.
    Cholesteryl esters in fish oils consisted mainly of C-16 and C-18 esters in fresh-water fish and C-14 to C-26 esters in salt-water fish, with emphasis on the longer chain length in the latter.

    The properties of hyaluronidase prepared from gastric mucosa of mackerel were studied. Determination of the activity in a 1:2 dilution of MCILVAINE buffer revealed an optimum pH of 4.8, which shifted to 2.8 with the addition of NaCl up to a concentration of 0.15M. Various chlorides such as NaCl, KCl and MgCl2 enhanced the activity, and their effect might be attributed to Cl-. At a concentration of 10-4 M Zn++ enhanced the activity, although Ag+ and Cu++ scarcely affected and Hg++ inhibited it. Polycation increased the activity; on the contrary, polyanion inhibited it. The maximum activity was found at approximately 50°C under the conditions tested. The MICHAELIS constant for potassium hyaluronate was 0.50mg per ml. The molecular weight was estimated to be approximately 56, 000 by the gel filtration method.

    The existence of thiaminase I in anchovy was verified by the fact that heteropyrithiamine was produced when the enzyme reaction was performed with thiamine hydrochloride as substrate added with pyridine. Also, some properties of the enzyme and its distribution in fish body were investigated. The results of the study are summarized as follows.
    1. The optimum pH value of the thiaminase is 5.5.
    2. The enzyme activity disappears completely when heated at 90°C for 10 minutes.
    3. The enzyme activity is inhibited by p-chloromercuribenzoic acid, iodoacetamide and monoiodoacetic acid but isrecovered up to 85% by cysteine.
    4. In regard to the distribution of thiaminase I in the fish body, the density is particularly high in the kidney, followed by the liver, pyloric caecum, intestine, stomach and muscle, respectively.
    5. The enzyme activity in anchovy as a whole is comparable to that in carp.
    6. The enzyme reaction is most activated by pyridine, followed by aniline and quinoline.

    In the examination various organic compounds, which would seem to be the growth factors of strains both CTC-sensitive and CTC-resistant or either strain of them, were tested in terms of the additives to the Minimal 10 (M 10) medium. The results obtained were as follows; 1) CTC-resistant strain grown in the M 10 medium was not affected by supplement of any single amino acid, but grew best on casamino acids. The growth of it was not accelerated only by B-vitamins, but also by various bases of nucleic acid; 2) On the contrary, the CTC-sensitive strain could not grow in the M 10 medium on supplement of either casamino acids, B-vitamins, or the combination of both components. However, it grow on purine bases of either adenine or of guanine. Additionally, the supplement of casamino acids and pyrimidine bases were more stimulative for its growth, 3) Comparatively growth of the CTC-sensitive strain in the medium fortified by the purine bases and the casamino acids were roughly identical to that of basal medium supplemented with yeast extract.

    A study was done on cofactors which may effect the activity of the β-oxidation in carp (Cyprinus carpio) hepatopancreas mitochondria. Finding that carnitine, CoA, ATP and Mg++ accelerated the 02 uptake, an improved medium was devised for the further study of the β-oxidation system.
    In the improved medium the 02 uptake was 4.7 times that of the original one, while the endogenous respiration remained almost unchanged. Removal of CoA and carnitine from the improved medium resulted in a considerable decrease of the 02 uptake suggesting that the activation of fatty acids and the permeation of the activated acids through the membrane are probably the initial stages of the β-oxidation phenomenon. Moreover, the 02 uptake in the improved medium was not affected until the sample mitochondria was left at 0°C for 12 hours.

    From the viewpoint of comparative biochemistry, the sterol compositions of mollusks belonging to the class Gastropoda (three orders, nine families, sixteen genera, seventeen species), Pelecypoda (two orders, three families, three genera, three species), and Polyplacophora (one species) were investigated by using gas-liquid chromatography on 1.5% OV-17.
    The gastropods were found to contain cholesterol as a main sterol. In the case of Murex asianus, Cymatiidae sp., Charonia sauliae, and Conomurex luhuanus, however, other C27- and C28- sterols were detected in addition to cholesterol (42-64%). Compared to the gastropods, the pelecypods appeared to contain relatively small amount of cholesterol (26-35%) and revealed the very complex composition of sterols. On the other hand, the chiton, Liolophura japonica, belonging to the most primitive mollusks, was found to contain 7-cholestenol ( ?? 7-sterol) contrasting that of the other mollusks.

    In the present study, the bioconversion of the dietary brassicasterol in Artemia salina was investigated. After administration of the Cyclotella nana containing brassicasterol-14C to Artemia, the sterols were isolated from the tissues of Artemia. The isolated sterol was found to contain only cholesterol and to reveal relatively high radioactivity. In column chromatography on a silver nitrate-impregnated silicic acid as acetate, cholesterol-14C gave one radioactive peak corresponding to cholesteryl acetate. Moreover, cholesteryl acetate-14C showed a constant specific activity (dpm/mg) during repeated crystallizations.
    On the basis of these results, it was concluded that Artemia is capable of converting the dietary brassicasterol to cholesterol.
