    The effect of transient local anoxia (10 to 120min) upon the endolymphatic potential (abbr. EP) was observed in 30 normal guinea pigs. The recovery of the EP was complete in the animal subjected to 10min anoxia and almost complete in 20min anoxia. When the duration of anoxia was 30min or longer, the recovery of the potential was incomplete, and the longer the duration of anoxia, the lower the level of the EP after reoxygenation. These results were discussed in relation to the possible role of a transient anoxia upon sudden deafness.

    In this report cerebro-cortical innervation of the urinary bladder has been investigated in man by the use of electroencephalography and in the cat by the application of the evoked potential technique. Electroencephalographic monitoring during bladder filling while the patient was asleep was a useful method to determine cerebral response to stimulation of bladder sensory receptors. Bladder filling was performed during stage IV sleep. In the intact individual detrusor reflex contraction or attainment of bladder capacity was followed by prompt arousal and desynchronization of EEG rhythms. In patients with interruption of bladder sensory pathways in the periphery or in the spinal cord there was loss of impairment of the EEG response to bladder filling. In the cat, anesthetized with sodium pentobarbital, electrical stimulation of the nerve supply to the urinary detrusor muscle and to the pudendal nerve innervation to the periurethral striated muscle and anal sphincter, evoked a diphasic response in the cerebral cortex of the frontal lobe. All responses were grouped in the same area and were of short latency. Similar potentials have been evoked in man. These studies demonstrate that the peripheral, spinal and cerebral innervation of the urinary bladder in man are definable by electrophysiologic techniques. As a result, these electrophysiologic techniques are applicable to defining neuropathic changes in the urinary bladder innervation in man.

    A new mass sreening method for detecting UDP-galactose-4-epimerase deficiency in newborn infants was devised so as to make a useful addition to the Guthrie test. Like Beutler's method, this method enables one to perform convenient spot tests on the basis of fluorometry of NADH. The preferred procedure was determined to be as follows: To a mixture of 10μl of 5-10 mM UDP-galactose, 10μl of 26mM NAD, 10μl of a 1 M tris-HCI buffer (pH 8.0) and 10μl of a 1/100 dilution of a 5mg/ml UDP-glucose dehydrogenase stock solution, distilled water is added until the total volume reaches 100μl. Then a blood-bearing filter paper disc is added to the mixture and, the reaction mixture is incubated at 37°C for 6-17hr. The resulting test solution is spotted on a Whatman No. 1 filter paper with a capillary tube, air-dried and tested for fluorescence with an ultraviolet light detector. No cases of UDP-galactose-4-epimerase deficiency were found in the present study.

    Crude glycosaminoglycans (GAG) separated from normal human urine by cetylpyridinium chloride-precipitation were fractionated with ethanol. The resulting fractions were then examined by electrophoresis on cellulose acetate membranes before and after enzymatic digestions and nitrous acid treatment. Analytical data for these fractions and the electrophoretograms sug-gested that ethanol-fractionation was useful to separate roughly chondroitin sulfate A, C-type GAG and heparan sulfate in human urine.

    Both (But)2cAMP and n-butyric acid suppressed the proliferation of TM-1 cells and induced the formation and elongation of the cellular processes in vitro. Prednisolone stimulated the proliferation of TM-1 cells and increased the number of polygonal cells in vitro. These results indicated that (But)2cAMP and n-butyric acid induce the differentiation of TM-1 cells, whereas prednisolone brings the cells in an undifferentiated state. The growth of TM-1 cell tumor transplanted into nude mice was suppressed by 5mg of (But)2cAMP and I mg of theophylline. Remarkable histological changes were seen in the tumors treated with these agents as compared with untreated tumors. This suggests that (But)2cAMP suppresses the proliferation of TM-1 cells in vivo.

    A new method was developed for the estimation of arginine vasopressin (AVP) in plasma and urine. Samples were extracted by a microcolumn of resin and assayed radioimmunologically using a highly sensitive antiserum to AVP. Ion-exchange resin, CG-50, H+ form, packed in a small column (diameter 4mm, height 6mm), was proved effective to remove the interfering substances and to concentrate the AVP in the sample. The application of 80% acid acetone successive to diluted HCl brought about a consistent recovery of AVP from the resin column. Recoveries were 66.4±8.5% for plasma and 85.4±9.7% for urine. In normal subjects plasma AVP levels were 3.9±0.3pg/ml (mean±S. D.) in ambulatory states, 4.9±0.6 after overnight fast, and 0.4±0.2 after water loading. High levels of 2.0-24.2pg/ml were obtained in patinents with syndrome of inappropriate secretion of ADH (SIADH), low values of 0-1.8 pg/ml in diabetes insipidus. Urinary excretions of AVP were 117.4±59.4ng/24hr (mean±s. D.) in normal controls, 191±177.0/24hr in SIADH, and 17.0±12.0/24hr in diabetes insipidus.

    Clinical and histopathological investigations were carried out in 71 cases of carcinoma of the corpus uteri treated at the Department of Obstetrics and Gynecology, Tohoku University Hospital over the 10 year period from 1968 through 1977. The ratio of cases of the corpus uteri to those of the cervix was 6.5/100. The average age of the former group was 58 years and 30% of them were nulligravida. The rates of accompanying hypertension and obesity were 21% and 20%, respectively. With regard to clinical stages, 76% were Stage I, 13% were Stage II, 10% were Stage III and 1% were Stage IV. The histological grade was G1 in 61%, G2 in 26%, and G3 in 13%. In Stage I, 80% of the cases had myometrial invasion into less than 1/3 of the uterine wall, but in Stage II invasion was limited to 1/3 in less than 25%. Metastasis to pelvic lymph nodes was found in 3 of 34 cases (9%): 1 of 24 cases of Stage I (4%) and 2 of 9 cases of Stage II (22%). All three of these cases had myometrial invasion reaching deeper than 1/3 of the uterine wall. Those with deep myometrial invasion showed a diffuse invasive pattern, as did those of G2 or G3 lesions. Cases of G1 lesions showed a relatively shallow myometrial invasion, whereas there was a tendency for cases of G2 or G3 lesions to have deep myometrial invasion. The over-all 5 year relative survival rate was 83%, while that for the Stage I patients was 98%. All 12 of the patients who died of recurrence of carcinoma of the corpus uteri died within 2 years of treatment.

    A 6-sulfated tetrasaccharide obtained by digesting chondroitin-6-sulfate with testicular hyaluronidase was used as a substrate for the determination of N-acetylgalactosamine-6-sulfate sulfatase activity. The activity was not detected in liver obtained from the elder sister with clinically classic Morquio syndrome and 4.7% of the control in liver from the younger sister with the same disease.

    An investigation was performed of the liver mitochondrial respiratory function in rats after 1, 3 and 6 weeks of biliary obstruction and the following results were obtained: 1) Various parameters of liver mitochondrial respiratory function, such as respiratory control ratio, ADP/O, O2 consumption in state 3 respiration and adenosine triphosphate synthesis were found to decrease with prolongation of biliary obstruction. 2) The mitochondrial respiratory enzymes, cytochrome a (+a3) and cytochrome c(+c1) showed that both decreased in contents with prolongation of obstruction, particularly the latter. 3) The activation ratio of ATPase (latent ATPase/dinitrophenol stimulated ATPase) was increased after long term biliary obstruction, which was thought to indicate the severe damage of mitochondrial membrane. 4) Investigation of the respiratory function with the various respiratory substrates showed that the locations of mitochondrial respiratory inhibition in obstructive jaundice are at sites 1 and 2, which is the same as the situation seen in nonspecific damage of mitochondria. 5) There was a high degree of mitochondria) respiratory disturbance by bile acids, particularly CDCA, which is thought to be one of the causal factors of liver dysfunction in obstructive jaundice. 6) Mitochondria] respiratory function was markedly disturbed in hypotension, and the degree of which correlated with length of time of biliary obstruction.

    The effects of bile from rabbits and deoxycholate on platelet aggregation and platelet morphology were studied. (1) Bile diluted to 1.25%, as well as 0.1% deoxycholate, significantly inhibited ADP-induced aggregation (p<0.01). (2) Ring-shaped platelets were observed in preparations treated with bile containing high concentrations of deoxycholate. These platelets lacked the center portion, but maintained the circumferential band of microtubules as revealed by scanning and transmission electron microscopy.

    A biochemical analysis of acidic glycosaminoglycans in cultured skin fibroblasts of a patient with epidermolysis bullosa acquisita was carried out. Acidic glycosaminoglycans were separated from fibroblasts and culture medium and subjected to quantitative and qualitative analysis by cellulose acetate membrane electrophoresis and column chromatography. Dermatan sulfate in acidic glycosaminoglycans was eluted out with 1.25M NaCI solution by Dowex 1 column chromatography. It was found that dermatan sulfate was increased in epidermolysis bullosa acquisita in comparison with the normal control. Then, the incorporation of 3H-glucosamine into cultured skin fibroblasts was investigated. Acidic glycosaminoglycans in the fibroblasts of the patient with epidermolysis bullosa acquisita were increased as compared with those of the normal control, although no significant differences were found in the metabolic patterns of acidic glycosaminoglycans between the patient and the normal control. Thus, it was thought that the increase in the amount of dermatan sulfate in the dermal tissue of the patient with epidermolysis bullosa acquisita was due to an excess production in the fibroblasts. It is, however, still obscure how this increase of acidic glycosaminoglycans in the dermis is related to the mechanism of the blister formation in epidermolysis bullosa acquisita.

    Serum gonadotropin response patterns to LH-RH infusions were investigated in women with and without actinomycin D treatment. LH and FSH levels increased until 60min in the nonovarectomized premenopausal women and until 120min in ovarectomized women after infusion in both groups. But, thereafter the levels in women untreated with actinomycin D were much higher than in women treated with actinomycin D. These results support the view that the LH-RH-stimulated -synthesis of gonadotropin involves increased RNA synthesis.

    SV40 temperature sensitive mutant transformed mouse cells synthesized different membrane glycoproteins at 5 to 10 hr after temperature shift and inhibition of DNA synthesis by cytosine arabinoside (Ara C) after temperature shift led to the alterations in the patterns of newly synthesized membrane glycoproteins analyzed by polyacrylamide gel electrophoresis.

    In order to study the metabolism of collagenous proteins in diabetes mellitus, urinary excretion of O-hydroxylysylglycosides, a characteristic component of collagenous protein, was determined in diabetics and controls. There was no significant correlation between urinary excretion rate of O-hydroxylysylglycosides and age (γ=-0.40 for diabetics and γ=-0.15 for control). In 16 age-matched pairs, urinary excretion of O-hydroxylysylglycosides was less in diabetics than in control (0.78±0.18 (mean±S. D.) μmoles/m2 body surface. hr versus 0.92±0.18 μmoles/m2 body surface-hr, p<0.05). This result suggests that catabolism of collagenous protein decreases in diabetics.

    Total fat balance was studied in 13 healthy controls and 14 patients with primary hyperlipidemia by estimating dietary and fecal fat. Although no difference in fat intake was noted between hyperlipidemics and controls, fecal fat excretion in hyperlipidemics increased significantly as compared with that in controls. The increase in fat excretion was greater in patients with type IIa hyperlipidemia (hypercholesterolemia) than in those with types lib and IV hyperlipidemia (endogenous hyperlipidemia).

    AmB/NCS combination therapy against cancer was evaluated. Six gastric cancer patients were treated in this manner, and 7 gastric cancer patients treated with NCS alone to serve as the control. AmB sirup was administered orally for 4 days before surgical operation. NCS was given intravenously at the onset of gastric surgery. Lesion tissues and healthy tissues were collected from each patient and the NCS titers were measured by bioassay. It was shown in the majority of the gastric cancer cases that the NCS levels in the lesion tissues were substantially higher than in the surrounding normal tissues, whereas in the gastric cancer patients who received NCS alone, no significant differences were found between the tissues.
