Protection of BALB/c Mice against Methicillin-Resistant Staphylococcus aureus Infection by Intraperitoneal Administration of Nucleoside-Nucleotide Mixture. Tohoku J. Exp. Med., 1993, 169 (3), 179-186- The effect of intraperitoneally administered nucleic acid components (nucleoside-nucleotide mixture) on the recovery from methicillin-resistant Staphylococcus aureus (MRSA) strain 8985N infection was studied in mice. Two experiments were conducted in which BALB/c mice were fed a nucleotide-free 20% casein diet for 30 days. On the 10th day, the mice were inoculated intravenously with viable MRSA organisms. The mice were intraperitoneally administered nucleoside-nucleotide mixture or saline (control) daily from the onset of the experiment (experiment 1) or from the day of inoculation (experiment 2). The survival rate in the nucleoside-nucleotide group in experiment 1 (70%) was significantly higher than that in the saline group (20%) (p<0.05). In experiment 2, the survival rate in the nucleoside-nucleotide group (55%) tended to be higher than that in the saline group (36%) without statistically significant difference; furthermore, in the surviving mice, the viable MRSA organisms recovered from the spleen and the kidney were fewer in the former group than in the latter group (p<0.05). The studies showed that the intraperitoneal administration either before or after the MRSA challenge of the nucleoside-nucleotide mixture was effective for the recovery of the mice from the infection.- MRSA; nucleoside; nucleotide; mouse

Prognostic Significance of Simultaneous Infiltration of HLA-DR-Positive Dendritic Cells and Tumor Infiltrating Lymphocytes into Human Esophageal Carcinoma. Tohoku J. Exp. Med., 1993, 169 (3), 187-195- The distribution of HLA-DR-positive dendritic cells (DR+DCs) and tumor infiltrating lymphocytes (TILs) was investigated in 67 patients with squamous cell carcinoma of the esophagus. DR+DCs were chiefly present among the cancer cell nests, and few were seen in the stroma. As clusters, UCHL-1-positive T-cells (UCHL1-Ts) were detected in the tissue around the cancer cell nests and as individual cells within tumor nests. Some UCHL1-Ts in the tumor nests showed direct contact with DR+DCs. In addition, a few OPD4-positive helper/inducer T cells were found among cancer cells, and were densely present in the stroma. A correlation was demonstrated between the number of DR+DCs and UCHL1-Ts in each patient (R=0.4547, p<0.01). In addition, dense tumor infiltration by both DR+DCs and UCHL1-Ts within the cancer cell nests was related to a better prognosis (p<0.01).- HLA-DR; dendritic cell; lymphocyte; esophageal cancer

    AMAKASU, H., KANNO, A. and ABE, M. Multiple Myeloma Occurring in Early Stage Primary Biliary Cirrhosis. Tohoku J. Exp. Med., 169 (3), 197-203- B-cell neoplasms are not infrequently associated with autoimmune diseases. A 60-year-old man with multiple myeloma developing in an early stage primary biliary cirrhosis was reported. Initially, he had hypergammaglobulinemia with monoclonal gammopathy (IgG-x type), elevation of serum IgG and IgM, and positive serum antimitochondrial antibodies. There were compression fractures of the lumbar vertebrae, where bone marrow aspiration revealed proliferating myeloma cells. Liver biopsy revealed destruction of bile duct surrounded by an inflammatory infiltrate, which was consistent with stage I primary biliary cirrhosis. The association suggested the role of immunoregulatory abnormalities in the development of multiple myeloma.- multiple myeloma; primary biliary cirrhosis; B cell neoplasm; immunological abnormality

Tohoku J. Exp. Med., 1993, 169 (3), 205-213- To assess the mechanism of insulin resistance in metabolic acidosis, we investigated insulin receptor binding and glucose uptake in isolated rat epididymal adipocytes at three pHs, acidic (pH 6.8), neutral (pH 7.4) and alkaline (pH 8.0) groups. After male Sprague-Dawley rats weighing 220-240g were submitted under chloral hydrate anesthesia, epididymal fat pads were dissected and isolated. [125I]-insulin binding rate was not different between alkaline and neutral groups, but that of acidic group was markedly decreased to approximately 30% of neutral group at maximum specific insulin binding rate. Scatchard analysis showed that numbers of insulin receptors were not different among three groups, but values of binding affinity in acidic group was significantly lower than those of the others. 2-[3H]deoxy-D-glucose uptake of alkaline and neutral groups were increased to reach steady state immediately, but that of acidic group increased slowly and reached at maximum which was lower than the other two groups. Furthermore, 2-deoxy-D-glucose uptake against amount of bound insulin was lower in acidic group than other two groups. From these results, it is suggested that insulin resistance in metabolic acidosis resulted from the combined defects in binding affinity and post-binding process in the insulin receptor.- insulin resistance; acidosis; insulin receptor; glucose uptake; adipocyte

    OHKUBO, M. Changes in 31P-MR Spectroscopy in Experimental Tumors Following Radiotherapy Combined with 5-Fluorouracil Compound (UFT). Tohoku J. Exp. Med., 1993, 169 (3), 215-223- The energy metabolism of FM3A tumors in C3H mice following radiotherapy (20 Gy or 40 Gy), 5-fluorouracil compound (UFT) chemotherapy or radiotherapy (20 Gy) combined with UFT was investigated by in vivo 31P-MR spectroscopy on day 2 after treatment. The effects were compared with the growth curves of the tumors and with the cytokinetic and histological changes in the tumors on day 5 after treatment. The combined group (20 Gy+UFT) showed growth curves similar to the 40 Gy group. Cytokinetic and histological changes of the combined group were also similar to those of the 40 Gy group, suggesting supra-additive effect of UFT. 31P-MR spectra of the combined group were similar to those of the 40 Gy group which were characterized by increase of intracellular pH(pHi), decrease of Pi/β-NTP and of PME/totalphosphorus -signal ratios. Spectra of the UFT group and the 20 Gy group, on the contrary, showed decrease of pHi, increase of Pi/β-NTP and of PME/totalphosphorus -signal ratios. Changes detected by 31P-MR spectroscopy on day 2 were considered to reflect early metabolic events which were correlated to the growth curve, cytokinetic and histological alterations observed on day 5 after treatment.- 31P-MRS; experimental tumor; irradiation; UFT

    CHECK, J.H., ZAVOS, P.M., KATSOFF, D., KIEFER, D. Effects of Percoll Discontinous Density Gradients vs SpermPrepTMII* vs Sephadex G-50 Gel Filtration on Semen Parameters. Tohoku J. Exp. Med., 1993, 169 (3), 225-231- There are various methods of separating sperm from seminal plasma for subsequent intrauterine insemination (IUI) and for in vitro fertilization (IVF). The purpose of the present study was to assess and compare semen parameters following Sephadex, Percoll and SpermPrepTMII separation techniques. The SpermPrepTM II is also a Sephadex preparation but uses a different bead size, less Sephadex and is a quicker method. The specimens (n=16) were initially evaluated for count, (C; ×106/ml) % motility (MO), grade of motility (GR; %), and HOS test scores. Each specimen was then divided into 3 equal aliquots and prepared as follows: 1) layered onto a modified Percoll discontinuous density gradient column; 2) processed using SpermPrepTMII; and 3) filtered through a Sephadex gel filtration column (G-50). The results show all 3 treatments yielded lower (p<0.01) counts as compared to control values but no differences were noted among them. Percoll and SpermPrepTMII increased MO (p<0.01) where Sephadex reduced it (p=NS). Percoll and SpermPrepTMII improved the % of best quality sperm but not Sephadex with SpermPrepTMII being higher than Percoll. There were increases (p< 0.05) in HOS values in all experimental treatments with SpermPrepTMII being the best. However, this study did not show as many males with HOS scores below 50% as noted in other studies. Finally, Percoll and SpermPrepTMII seem equally effective methods for producing high quality sperm for IUI or IVF although SpermPrepTMII is quite faster.- SpermPrepTMII; Sephadex; Percoll discontinuous density gradient; hypoosmotic swelling test; semen filtration

Tohoku J. Exp. Med., 1993, 169 (3), 233-244- Pulmonary hemodynamics of Bleomycin-induced interstitial fibrosis model (group-BLM, n=10) and saline-treated control (group-C, n=12) were studied in awake rats. Four weeks before hemodynamic study, bleomycin sulfate and normal saline was intratracheally instilled to the group- BLM and the group-C, respectively. Pulmonary artery and abdominal aortic catheters were indwelled two days before the hemodynamic study. In room air, mean pulmonary artery pressure (Ppa) was significantly higher and systemic artery pressure was significantly lower in group-BLM than in group-C; Ppa=21±1 cmH2O (mean±S.E.) for group-C and 38±4cmH2O for group-BLM. Cardiac output did not differ among the groups. Mean pulmonary vascular resistance (PVR) of group-BLM was double that of group-C. Right ventricle was hypertrophic in group-BLM. When exposed to 10% O2, PVR of group-C significantly rose, whereas PVR of group-BLM showed very little increase, showing attenuation of the hypoxic pulmonary vasoconstriction (HPV). Magnitude of the HPV was inversely related to Ppa during air breathing. We conclude that notable pulmonary hypertension and right ventricular hypertrophy occur, and HPV is blunted four weeks after bleomycin instillation, at the most severe period of this lung fibrosis model. We speculate that the high intravascular pressure partly contributed to the blunted HPV in bleomycin-treated group.-bleomycin; pulmonary interstitial fibrosis; pulmonary hypertension; hypoxic pulmonary vasoconstriction

Female Systemic Lupus Erythematosus in Miyagi Prefecture, Japan: A Case-Control Study of Dietary and Reproductive Factors. Tohoku J. Exp. Med., 1993, 169 (3), 245-252- To investigate risk factors of systemic lupus erythematosus (SLE) in relation to diet and reproduction, a populationbased case-control study was conducted during the period from October 1988 to October 1989 in Miyagi Prefecture in northeastern Japan. Included in the study were 52 female patients with the recent SLE onset. Two sex- and birth yearmatched (±2 years) controls for each patient were selected from the general population. The analysis on diet showed that the frequent intake of meat was associated with an increased risk (frequent vs. rare, relative risk (RR) 3.36; 95% confidence interval (CI) 1.10-10.24) and that the patients preferred fatty meat such as beef or pork. The analysis on menstrual history revealed that menstrual irregularity was also associated with an increased risk (RR 3.79; 95% CI 1.43- 10.01). These results suggest that dietary and reproductive factors may be responsible for the onset and the progression of SLE.- case-control study; diet; reproduction; risk factor; systemic lupus erythematosus

Med., 1993, 169 (3), 253-255- To attain its increased tumor concentration and to avert its systemic adverse effects, aerosol cisplatin (CDDP) was incorporated into radiotherapy (RT) of laryngeal cancer. Nine patients were asked to inhale the aerosol following each RT session. Their clinical tumor response was favorable, and histopathologic survey in selected cases revealed elimination of viable cancer cells. However limited, there have been no reports of cancer recurrence yet. This reinforcing plan of aerosol CDDP for RT bleomycin would certainly offer a better way of treating laryngeal cancer, and probably those malignancies facing the nasopharyngobronchial airway as well.- aerosol cisplatin; bleomycin; laryngeal cancer; radiotherapy

Rises in Urinary Bicarbonate Contents and pH of Adult Tokyo Citizens. Tohoku J. Exp. Med., 1993, 169 (3), 257-259- Urinary bicarbonate has been considered to be negligible, and the pH of urine to be low (around pH 5.5) for these fifty years. It was, however, found that the contents of bicarbonate and pH of the urine of adult Tokyo citizens (435 adults including 148 males and 287 females) substantially increased. The findings may be correlated with increased inhaled amounts of carbon dioxide which finally causes the increase of urinary excretion of bicarbonate, though evidence that the gas accumulates in the atmosphere of Tokyo due to air pollution has not been directly demonstrated.- urinary bicarbonate; urinary pH; atmospheric carbon dioxide
