    YAMADA, S., SASANO, N. and NAKAMURA, T. Immunohistochemical Localization of Brain-Gut Hormones in Gastric Carcinoma with Relation to Argyrophil Cells. Tohoku J. exp. Med., 1984, 143 (1), 1-15 - A total of 87 surgical cases of gastric carcinoma including 3 carcinoid tumors were investigated with the methods of silver reaction and immunoperoxidase stain for 8 different brain-gut hormones. Argyrophil (AP) cells were demonstrated in 38 cases (44%), argentaffin (AF) cells in 18 (21%) and endocrine cells in 13 (14%). The occurrence of endocrine cells had no relation with histological types. Glicentin cells were demonstrated in 10 cases, somatostatin in 7, motilin in 3, β-endorphin in 2 and gastrin in one. Endocrine cells appeared generally in small numbers except one carcinoid tumor which had numerous somatostatin cells. No single cell positive for more than two kinds of hormones could be demonstrated. Two undifferentiated carcinomas looking like carcinoid tumors had argyrophil cells and endocrine cells of either somatostatin or β-endorphin. These results suggest that carcinoid-like carcinoma or endocrine cell carcinoma may lie on the intermediate state between carcinoma and carcinoid tumor, -gastric carcinoma; carcinoid tumor; argyrophil cell; endocrine cell; brain-gut hormone

Effect of Meal and Cimetidine on Endogenous Plasma Gastrin, Secretin and Pancreatic Polypeptide in Experimental Acid Hypersecretion in Dogs. Tohoku J. exp. Med., 1984, 143 (1), 17-25-In order to observe the plasma gastrin, secretin and pancreatic polypeptide (PP) levels in an acid hypersecretion state, the pyloric antrum was transposed onto the transverse colon in 4 mongrel dogs with a Heidenhain pouch. The effects of meal and cimetidine on these gut hormones and acid secretion levels were observed for 6hr after meals. After antrocolic transposition (ACT), hypersecretion of acid and high plasma gastrin levels were observed both in the fasting and stimulated states. Plasma secretin levels were elevated for 6 postprandial hr, but were reduced by cimetidine as a result of marked suppression of acid secretion. Plasma PP levels also increased for 6 postprandial hr, but its responses were not altered with or without simultaneous administration of cimetidine. These findings indicate that acid hypersecretion in basal and postprandial states after ACT may be attributed to hypergastrinemia and that endogenous acid is capable of releasing secretin but not effective on the release of PP.-gastric acid; intraduodenal pH; gastrin; secretin; pancreatic polypeptide

    CRIBA, T. and BOLES, E.T., Jr. Studies on the Relationship between the Number of Arteriae Rectae of Intestinal Artery and Intestinal Length. Tohoku J. exp. Med., 1984, 143(1), 27-31-In the measurement of the length of preserved intestine after massive resection of the intestine, measurement of the length of intestine as it is at the time of operation may produce a large error when compared to the length of the intestine at normal times. Therefore, attention was paid to the number of arteriae rectae of the intestinal artery and a comparison was made between such number and the length of the intestine at normal times in rats and man. As a result, the conclusion was drawn that the measurement of arteriae rectae of the intestinal artery is useful for the estimation of preserved intestine at normal times.-intestinal artery; arteriae rectae; intestinal length

    KOBARI, M., MATSUNO, S., SATO, T., KAN, M. and TACHIBANA, T. Establishment of a Human Pancreatic Cancer Cell Line and Detection of Pancreatic Cancer Associated Antigen. Tohoku J. exp. Med., 1984, 143 (1), 33-46-We succeeded in an establishment of a human pancreatic cancer cell line (PK-1) from liver metastasis of pancreatic cancer. Primary pancreatic cancer cells grew as islands surrounded by fibroblastic cells. However, these fibroblastic cells were gradually omitted by the polygonal shaped cancer cells. This cell line contained neither zymogen granules nor trypsin indicating that this pancreatic cancer originated from pancreatic duct cells. Modal chromosome numbers of this cell line were 42 and 72 and the doubling time was 48hr. This cell line was transplantable in athymic nude mice to form progressive tumors which had histology similar to that of the original cancer (papillotubular adenocarcinoma). Neither AFP nor ferritin but CEA was detected on the surface and in the cytoplasm of this cell line in indirect immunofluorescence. Rabbit antiserum against this pancreatic cancer cell line detected pancreatic cancer associated antigen besides CEA in the culture supernatant. This antiserum reacted with sera from patients with pancreatic cancer to form a distinct precipitin line in agarose gel which fused with the precipitin line formed between the culture supernatant of this cell line and the antiserum.-pancreatic cancer cell line; CEA; transplantable tumor; pancreatic cancer associated antigen

Changes in Placental Lactogen, Beta- Chorionic Gonadotropin, and Unconjugated and Total Estriol Levels in the Late Course of Normal Human Pregnancy. Tohoku J. exp. Med., 1984, 143 (1), 47-51-The levels of human placental lactogen (hPL), beta-human chorionic gonadotropin (β-hCG), unconjugated estriol (UE3) and total estriol (TE3) were measured radioimmunologically in women along their course of pregnancy. The serum hPL and β-hCG levels were relatively constant in uncomplicated pregnancy, while UE3 and TE3 significantly rose towards 41 weeks of gestation. There were positive correlations between hPL and β-hCG (n=49, r=0.737, p<0.001), and between UE3 and TE3 (n=49, r=0. 904, p<0.0001). The ratios of placental hormones to estriol showed moderate declines towards 41 weeks of gestation. These data suggest that the fetal adrenal function increased toward the term, while the placental peptide hormones decrease or remain unchanged. Determination of serum unconjugated estriol was found to be convenient and useful for monitoring the fetoplacental function.-hPL; beta-human chorionic gonadotropin; unconjugated estriol; total estriol; pregnancy

    IIOKA, Y., KIKUCHI, K., TADA, H., ISOGAI, S. and URAYAMA, T. Plasma Soluble Fibrin Monomer Complexes in Nephrotic Syndrome-with Reference to Hypoalbuminemia. Tohoku J. exp. Med., 1984, 143(1) 53-57-Plasma soluble fibrin monomer complexes (SFMC) in 10 patients with nephrotic syndrome were measured to demonstrate the contribution of hypoalbuminemia to the SFMC formation. The levels of SFMC as well as plasma fibrinogen (Fbg) levels were significantly higher than those in control subjects. There was a negative correlation between the levels of SFMC and serum albumin, and also between Fbg and serum albumin. The increase in SFMC levels which indicates the intravascular generation of thrombin might be correlated to hypoalbuminemia in nephrotic syndrome with hypercoagulability.-soluble fibrin monomer complexes; hypoalbuminemia; nephrotic syndrome; hypercoagulability

    HASHIMOTO, K., KAMITANI, T., WADA, Y. and TATSUMI, N. Presence of Connectin-like Protein in White Blood Cells and Platelets. Tohoku J. exp. Med., 1984, 143 (1), 59-70-Connectin is known to be an anchoring protein of actomyosin filaments in skeletal muscle. Attempts were made to extract this protein from white blood cells and platelets in order to investigate its properties. The purified connectin-like protein obtained had a rubbery appearance and was insoluble in water and immobile on SDS gel-electrophoresis. Amino acid composition and electron microscopic features of the protein were similar to those of skeletal muscles. FITC-labeled antibody to the protein showed. a positive reaction to the membrane fractions of red cells, white cells and platelets. In moving cells fluorescence was observed not only in pseudopod, but also in whole cytoplasm. No changes in the fluorescence patterns were observed with respect to moving stages or cell types. The presence of the protein beneath the cell membrane was also confirmed electron micrographically.-connectin-like protein; platelets; leukocytes

    MAEBASHI, M., YOSHINAGA, K. and Suzuki, M. Effect of Pindolol, a β-Adrenoceptor Blocking Agent, on Lipid Metabolism. Tohoku J. exp. Med., 1984, 143 (1), 71-77-Pindolol, a β-adrenoceptor blocking agent, was administered to rats to determine the effects of the drug on lipid and lipoprotein in plasma and tissues. The drug caused significant reductions in plasma triglyceride, free fatty acid and very low density lipoprotein. As for lipoprotein lipase activity, the drug produced varying effects ; an increase in the heart and a reduction in the adipose tissue. No significant changes in plasma cholesterol, low density lipoprotein and high density lipoprotein cholesterol were observed. Med., 1984, 143 (1), 79-85-The histamine level of whole blood and plasma in asthmatic patients was estimated by means of a simple, sensitive and specific method, which was developed to measure low histamine levels. This method consists of the following procedures; the partial purification of histamine with a small P-cellulose column; its further purification with high performance liquid chromatography (HPLC); and fluorometric detection with precolumn o-phthaldehyde (OPT) reaction. The present assay could detect as little as 0.5ng of histamine concentration. Blood histamine levels in patient with asthmatic attack, 57±34ng/ml (mean±S.D. N=14), were significantly different from those in symptom-free period, 37±15ng/ml (N=15) as well as those in normal subjects, 36±17ng/ml (N=12). However, there were no significant differences among plasma histamine levels in normal subjects, 1.6±1.7 ng/ml (N=12) and, in asthmatic patients during attacks, 1.6±1.8ng/ml (N=14) and symptom-free periods, 1.6±1.7ng/ml (N=15). These results indicate that plasma histamine concentration does not increase during asthmatic attacks, even though there was significant increase of blood histamine concentration.-histamine; HPLC; bronchial asthma

Med., 1984, 143 (1), 87-92-Serum prolactin and estradiol-17β levels were determined in umbilical cord sera obtained from 33 preterm, 118 full-term and 11 anencephalic infants and in peripheral sera obtained from 5 neonates. Prolactin levels increased from mid-gestation toward term, and markedly decreased after birth. Those in anencephalic infants (253±86ng/ml, mean±S.D.) were lower than in full-term infants (324±119ng/ml), but the difference was not statistically significant. We also performed thyroid-releasing-hormone (TRH) (50-200μg) test on 5 anencephalic infants, but significant changes of serum prolactin levels were not found. Estradiol-17β levels in anencephalic infants were significantly lower than those in full-term infants. There was a significant positive correlation between cord prolactin and estradiol-17β levels in full-term infants. These results suggest one of the control factors for human fetal pituitary prolactin secretion is estrogen, and hypothalamic factors are not so important.-prolactin; estradiol; TRH; fetus

Med., 1984, 143 (1), 93-98-In 3 patients with Zollinger-Ellison syndrome, gastric pepsin and acid secretion, and pepsin acid correlation were studied. The results obtained are as follows. In nocturnal secretion, and histamine-, insulin- and gastrin-stimulated secretion, pepsin hypersecretion was not as conspicuous as acid hypersecretion, and so pepsin acid correlation showed low levels of less than 1.0. Not only before operation but also after vagotomy or distal gastrectomy, the same results were obtained. These results suggest that both before and after operation Zollinger-Ellison syndrome should be suspected in patients with pepsin acid correlation of less than 1.0 in nocturnal secretion or stimulated secretion.-Zollinger-Ellison syndrome; pepsin secretion; acid secretion; pepsin acid correlation; vagotomy; distal gastrectomy

Med., 1984, 143 (1), 99-105-Agglutinability of red blood cells against anti-D increased remarkably when the cells were treated with proteolytic enzymes, such as bromelain, chymotrypsin, ficin, papain, pronase and trypsin. When Stroma prepared from normal red blood cells was treated with any of proteolytic enzymes, however, Rh-Hr blood type activities were completely abolished. The similar results were obtained from stroma solubilized with detergents which was treated with enzymes after being prepared. Of all the enzymes, ficin acted more slowly than the others did. Neuraminidase or phospholipase A2 had no effect on Rh-Hr activities at all. SDS-polyacrylamide gel electrophoretic pattern of stroma prepared from bromelain, ficin and pronase-treated red blood cells were quite different from that of normal stroma.-Rh-Hr blood type; proteolytic enzymes; red blood cell stroma

Med. 1984, 143 (1), 107-112-Intima-media of porcine, canine, rabbit, and human thoracic aortas were separately digested with pronase, after extraction of fat. Glycosaminoglycan fractions were then separated from the resulting glycoconjugate fractions by precipitation with cetylpyridinium chloride. Glycosaminoglycan compositions were determined by sequential digestion with Streptomyces hyaluronidase, chondroitinase AC, chondroitinase ABC and heparitinase 1. All the tissues contained hyaluronic acid, chondroitin sulfates AC, dermatan sulfate and heparan sulfate. Of these glycosaminoglycans, chondroitin sulfates AC were the most prominent ones. Relatively large proportions of heparan sulfate were contained in these tissues except for canine aorta. It is noteworthy that the proportions of glycosaminoglycans in the intima-media of rabbit and human thoracic aortas with similar maturation were quite similar, although those of others differed from each other.-glycosaminoglycan; thoracic aorta; aortic glycosaminoglycans

Med., 1984, 143(1), 113-116-Intima-media of thoracic aortas obtained from estrogen-treated, estrogen-progesterone-treated and sham-treated ovariectomized rabbits were separately digested with pronase after extraction of fat. Glycosaminoglycan fractions were then separated from the resulting glycoconjugate fractions by precipitation with cetylpyridinium chloride. The glycosaminoglycan levels increased after treatments with estrogen and estrogen-progesterone. Glycosaminoglycan compositions were determined by sequential digestion with Streptomyces hyaluronidase, chondroitinase AC, chondroitinase ABC and heparitinase 1. The proportions of dermatan sulfate and chondroitin sulfates AC increased after treatments with these hormones, while those of hyaluronic acid and heparan sulfate decreased. The changes of the proportions of dermatan sulfate and hyaluronic acid were dramatic. Med., 1984, 143(1), 117-124-To examine common antigens between human and monkey sera, sera from 19 kinds of monkey were investigated with crossed and rocket immunoelectrophoresis using antisera to human serum proteins. In the crossed immunoelectrophoresis, the number of precipitation lines, 40 in human and 8 in the lower primates, increased according to the order of phylogenetic classification, and the lines due to albumin and γ-globulin were observed in all the monkey sera examined. Quantitative investigation was performed by rocket immunoelectrophoresis with 8 kinds of mono-specific anti-serum protein. α2-Macroglobulin, orosomucoid, transferrin, IgG and IgM were not observed in monkeys of Loricidae family. Relatively large amounts of orosomucoid, albumin, IgG and transferrin were detected in monkeys of Cebidae family. It was considered that the distribution of serum proteins possessing antigens common to human is not related with the order of phylogenetic classification.-species-specificity; common antigen; serum proteins

    KOYAMA, K., GOTO, H., OUCHI, K. and SATO, T. Nuclear DNA Pattern of the Gallbladder Cancer. Tohoku J. exp. Med., 1984, 143(1), 125-126-Nuclear DNA of the gallbladder cancer was stained by the Feulgen method and analyzed using a new concept of DNA score. DNA score was determined by the peak ploidy unit and decided numbers of frequency in each ploidy unit. As the result, a significant correlation between DNA score and the depth of cancer invasion was demonstrated. The depth of cancer invasion, as reported already, has close correlation to the prognosis of the patients of gallbladder cancer. These results indicate that the nuclear DNA analysis is useful for the determination of malignancy and the prediction of the prognosis of the patients.-gallbladder cancer; nuclear DNA; depth of cancer invasion; Feulgen staining

    MIZUNO, K., GOTOH, M. and FUKUCHI, S. Acute Effect of Captopril on Serum Lipid Peroxides Level in Hypertensive Patients. Tohoku J. exp. Med., 1984, 143 (1), 127-128-Captopril, an orally active angiotensin-converting enzyme inhibitor, was administered to 15 patients with essential hypertension. The serum lipid peroxides level, aldosterone concentration in plasma and blood pressure decreased rapidly after administration, while plasma renin activity was not significantly changed. It is suggested that inhibition of angiotensin-converting enzyme by captopril offers a possible therapeutic approach to the treatment of atherosclerosis complicated with hypertension.-lipid peroxides; captopril; renin-angiotensin-aldosterone system; hypertension
