Med. 1984, 142 (4), 351-361-Mouse Clq, a subcomponent of the first component of complement, was purified from mouse EDTA plasma by a combination of precipitation with polyethyleneglycol, affinity chromatography on IgG-Sepharose, ion exchange chromatography and molecular sieving. Mouse Clq was compatible with human Clq in the sense that it shows Cl hemolytic activity by the combination with human Clr and Cls. The molecular weight of mouse Clq as estimated by SDS-PAGE was approximately 410, 000 and almost the same as that of human Clq. After treatment of mouse Clq with 4M urea, three distinct subunits were obtained on SDS-PAGE in non-reducing condition. Moreover it was shown that there are small, yet definite differences in the molecular weights of the subunits between the two mouse strains. Three subunits of the Clq from C3H and ICR showed apparent molecular weights of 62, 000, 58, 000 and 53, 000 but those of DDI showed 62, 000, 55, 000 and 51, 000. These results suggested the existence of allotype of mouse Clq, and indeed, alloantisera made by injecting the Clq of ICR to DDI reacted in Ouchterlony double diffusion test with the plasma of ICR, C3H, AKR and BALB/c, but not with DDD, DKI, C57BL/10 and C57BL/6. -mouse Clq; subunits of mouse Clq; allotype of mouse Clq

Med., 1984, 142 (4), 363-379-We carried out our Systematic Rhythm Test for the Brain-Damaged Patients named “The Asian Rhythm Test” on our four patients with lesions of the corpus callosum, and analyzed their rhythmic capabilities-their performances in rhythm perception (auditory distinction) and in rhythm expression (rhythm-tapping)-in accordance with the neuropsychological model proposed in 1980 by us. All our partial split-brain patients showed the abnormal one-ear inferiority (superiority) and the one-hand inferiority under our testing conditions. The patients with lesions of the anterior corpus callosum showed the interhemispheric (the left-to-right) transfer-dysfunction of motor-programming information, while the patient with the lesion of the posterior corpus callosum showed the interhemispheric transfer-dysfunction of auditory information. The patients with lesions including the truncus showed the interhemispheric integration-loss in the process of rhythm perception and its expression. And moreover, we could see the specific auditory interruption between the ipsilateral and the contralateral systems in the patient with the lesion including the anterior commissure. Med., 1984, 142 (4), 381-389-A rabbit model of acute hypersensitivity pneumonitis was produced by immunization and subsequent inhalation of pigeon serum. The pathological features, consisting of alveolitis and interstitial granulomas, were similar to those reported previously in the experimental models. Repeated inhalation challenge of pigeon serum, however, did not develop chronic alveolitis but led to the fading of the disease. It was suggested that desensitization defined as the suppression of delayed hypersensitivity contributed to the mechanism of the fading of the disease. Repeated pigeon serum inhalation without previous immunization by the same antigen could produce a moderate interstitial pneumonitis but did not demonstrate a typical granulomatous histological change in lungs. It was necessary to use adjuvant for producing a complete animal model of hypersensitivity pneumonitis. Antigen-specific antibody response did not directly correlate with the development of the animal model of hypersensitivity pneumonitis. -hypersensitivity pneumonitis; pigeon breeder's lung; rabbit model

Tohoku J. exp., 1984, 142 (4), 391-398 - A new method of examination whereby urethral pressure profiles are recorded from the anterior, right, posterior and left urethral walls and tomographically expressed has been devised. The system facilitates three-dimensional delineation of the functional morphology of the urethra, requires no more time and work than dose the conventional urethral pressure profile study, or causes no pain in the patient. Stereographic depiction of the continence mechanism of the bladder neck and the function of the external urethral sphincter was achieved by the method, and the degree of urethral oppression was assessed by application of this urethral pressure tomography in patients with prostatic hypertrophy.-urethral pressure profile; three-dimensional expresion

Serum Erythropoietin (ESF) Levels and Erythroid Progenitors (CPU-Es) of Patients with Chronic Myeloproliferative Disorders. Tohoku J. exp. Med., 1984, 142 (4), 399-407 - Serum erythropoietin (ESF) levels and the numbers of marrow and blood erythroid progenitors (CFU-Es) of patients with chronic myeloproliferative disorders (CMPD) were studied simultaneously. The numbers of marrow and blood CFU-Es per 1×105 cells were normal or greatly elevated. There was an inverse correlation between the hemoglobin concentration and the serum ESF level in patients with chronic myelogenous leukemia when the hemoglobin concentration ranged from 9.0 to 13.0g/100 ml. The serum ESF level was closely related to the hemoglobin concentration in CMPD and it was suggested that the negative feedback mechanism might operate in anemic patients with CMPD. - chronic myeloproliferative disorders; fetal mouse liver cell assay; erythroid progenitor (CFU-E); erythropoietin (ESF)

    OHKAWA, H. Effects of ATP, Its Related Nucleotides and VIP on the Inhibitory Potentials in the Smooth Muscle Cells of the Guinea-Pig Duodenum. Tohoku J. exp. Med., 1984, 142 (4), 409-422 - Non-adrenergic inhibitory potentials (i.p.'s) were recorded from the smooth muscle cells of the guinea-pig duodenum following field stimulation. ATP, ADP and AMP increased the membrane potential of the cell and decreased the amplitude and the rate of hyperpolarization of the i.p. The amplitudes of the maximum hyperpolarization due to a repeated stimulation and the post-stimulus depolarization were reduced by ATP and AMP. Methylxanthine could not inhibit the i.p. while apamin reversibly decreased the amplitudes of the i.p. and the post-stimulus depolarization. The latency and the time to peak were prolonged by apamin. Biphasic effect of quinidine on the i.p. was obtained. The amplitude and the rate of hyperpolarization of the i.p. were decreased at 10-4g/ml of quinidine. The amplitudes of the maximum hyperpolarization by repetitive stimulation and the post-stimulus depolarization were decreased by quinidine. The membrane potential of the cell was not affected by VIP. The amplitude and the other parameters on the i.p. were not changed in the used concentrations of VIP. Trypsin reduced the membrane potential of the cell but not changed the parameters on the i.p. The membrane was slightly depolarized by the treatment with papain while no significant changes on the i.p. were observed in papain. The obtained results suggest that ATP, its related substances and VIP do not contribute to the non-adrenergic inhibitory neurotransmission. -non-adrenergic transmission; inhibitory potential; ATP; VIP; duodenal smooth muscle

Med., 1984, 142 (4), 423-426-Immunoreactive ovine corticotropin-releasing factor (CRF) was revealed in the cell bodies of parvocellular neurons in the paraventricular nucleus of the human hypothalamus by an immunocytochemical technique. The immunoreactivity was negative in magnocellular neurons. Immunoreactive nerve fibers were found in the subependymal layers of the third ventricle and in the perivascular space of the primary plexus in the lower hypothalamus.     TAMAI, M. and MIZUNO, K. Distribution of Intra- and Extraocular Pain Induced by Argon Laser Photocoagulation. Tohoku J. exp. Med., 1984, 142 (4), 427-435 - We examined the characteristics of ocular pain induced by green argon laser photocoagulation in 13 eyes of ten patients. The patients had retinal detachment or proliferative diabetic retinopathy treated surgically by a 360 degree buckling procedure or vitrectomy with an encircling band on the equator before the laser treatment. All patients complained of ocular pain in various degrees during coagulation on and anterior to the buckle. We confirmed that this kind of ocular pain was asymmetrically distributed; it was dense in the superotemporal quadrant, sparse in the inferonasal quadrant, and intermediate in the remaining two. Seven patients (eight eyes) complained not only of ocular pain, but also of extraocular pain in the ipsilateral upper lid, forehead, and parietal and temporal areas of the head. Both kinds of pain diminished by retrobulbar anesthesia, or by the time interval following surgery. - ocular pain; argon laser photocoagulation; referred pain; ocular surgery; scleral buckling procedure

Tohoku J. exp. Med., 1984, 142 (4), 437-443 - The palmar aponeurosis of patients affected with Dupuytren's disease was examinded for collagen characteristics with regard to extractability, polymorphism, and posttranslational modifications, and the results were compared with those from normal subjects. The increased proportion of type III collagen relative to type I collagen in the affected tissue confirmed the previous findings in this disease. A slight but significant increase in a ratio of glucosylgalactosylhydroxylysine to galactosylhydroxylysine in the Dupuytren's tissue may be interpreted by the increase in the content of type III collagen. The affected tissue contained increased amounts of dihydroxylysinonorleucine as the reducible cross-link of collagen. These data support the view that Dupuytren's tissue contains collagen resembling that in granulation and embryonic tissues. Pyridinoline was shown to occur in normal and affected aponeurosis. No change in its content suggests that this cross-link is not involved in the pathogenesis of contracture in this disease.-collagen; aponeurosis; Dupuytren's disease; pyridinoline; hydroxylysine

Med., 1984, 142 (4), 445-452 -The role of endogenous dopamine (DA) on plasma GH secretion was studied in 12 patients with active acromegaly. After intramuscular administration of 100mg sulpiride, 3 out of 12 patients showed distinct increases in their plasma GH (cases 1-3: sulpiride- responders). The GH increases far exceeded the ranges of physiological fluctuation in two cases studied (case 1 & 3). After domperidone administration, case 3 showed a quite similar GH increase as after sulpiride. Plasma GH responses to TRH and sulpiride were still observed in case 2 even after the basal GH levels were normalized by the transsphenoidal hypophysectomy. The three sulpiride-responders showed so-called paradoxical GH decreases following the administration of L-dopa (500mg, p.o.) or DA (5μg/kg/min over 90min, i.v.). However, the basal plasma PRL levels and the responses to sulpiride in these 3 cases were not homogeneous. These results indicate that endogenous DA has a tonic inhibitory effect on GH secretion in some cases of acromegaly, and that tumorous somatotrophs rather than elevated GH levels are the critical factor for such inhibition. Further, the sensitivities of somatotrophs and lactotrophs to DA appeared to be heterogeneous in acromegalic patients. - acromegaly; GH; sulpiride; domperidone; dopamine; TRH

Determination of Low Affinity Platelet Factor 4 in Frozen and Thawed Human Platelets by the Newly Developed Enzyme Immunoassay System. Tohoku J. exp. Med., 1984, 142 (4), 453-460 - By the use of a newly developed sandwich enzyme immunoassay method for low affinity platelet factor 4 (LA-PF4), the effects of repeated freeze-thawing on the contents of this protein in platelets were determined and compared with those of Triton-X 100 lysed platelets. The assay system consisted of polystyrene balls covered with immobilized antibody fragments F(ab')2 and the same antibody Fab' labeled with β-D-galactosidase from E. coli. The assay was specific to LA-PF4 with no significant cross-reactivity with platelet factor 4. Coefficients of variation within-run and between-run for the assay of LA-PF4 were less than 12%. The results obtained with this enzyme immunoassay correlated well with those of a radioimmunoassay of β-thromboglobulin which is immunologically identical with LA-PF4 (γ=0.961, slope=1.056, y-intercept=-3.739ng/ml; n=22). The contents of LA-PF4 per 109 Triton-X 100 lysed platelets in platelet-rich plasma were 23.65±3.14μg (mean±S.D.). The contents of LA-PF4 in platelets were increased from 46% to 95% of Triton-X 100 values by repeated freezing and thawing 1 to 7 times. The present data indicate that the freeze-thawing technique should be done carefully to obtain the reliable determination of LA-PF4 in platelets. - low affinity platelet factor 4; enzyme immunoassay; intraplatelet content; freeze thawing

    KIMURA, I. and AYYAR, D.R. Sensory Nerve Conduction Study in the Medial Antebrachial Cutaneous Nerve. Tohoku J. exp. Med., 1984, 142 (4), 461-466-Sensory nerve conduction in the medial antebrachial cutaneous nerve was studied in 40 normal subjects utilizing both antidromic and orthodromic methods. Using the antidromic method the distal sensory latency to the take-off point of the sensory action potential (SNAP) was 1.54±0.17msec, latency to the peak of the SNAP was 2.11±0.16mse, the amplitude of the SNAP was 18.8±7.1μv and the nerve conduction velocity was 65.9m/sec. Using the orthodromic method the values were 1.55±0.17msec, 2.10±0.15msec, 15.7±6.3μv and 65.5±8.5m/sec, respectively. Amplitude of the medial antebrachial cutaneous SNAP was usually smaller than that of the lateral antebrachial cutaneous nerve (p<0.01). This technique should be useful in the electrodiagnostic differentiation between eighth cervical or first thoracic root involvement and postganglionic lesions including the lesions of the inferior trunk or the medial cord of the brachial plexus and also in confirming involvement of this peripheral nerve due to other focal causes.-medial antebrachial cutaneous nerve; lateral antebrachial cutaneous nerve; sensory nerve conduction; electrodiagnosis

    MIYAGAWA, I. Urethral Pressure Changes during Voiding. Tohoku J. exp. Med., 1984, 142 (4), 467-468 - Intraurethral pressure changes in the external sphincter zone during voiding were recorded using a specially constructed double lumen urethral catheter in 3 normal subjects. It was noted in all the 3 subjects that the urethral pressure in this zone dropped simultaneously with decrease of electromyographic activities of the external urethral sphincter when the subjects strained to void. It was ascertained for the catheter position to remain unchanged irrespective of voiding action during urethrometrography. -urethral pressure change; voiding
