MN blood grouping of minute blood stains has been performed by various methods, but the results are not satisfactory. In the present experiments, various kinds of anti-M and -N sera such as non-adsorbed, adsorbed, and neutralized antisera and immunoglobulin class antibodies fractionated by DEAE-cellulose were applied to the group-specific double combination method (GSDCM). Minimum titers to induce the agglutination ranged from 1:1 to 1:2 in non-adsorbed antisera and from 1:64 to 1:128 in adsorbed ones. Double combinating potency (DCP) of non-adsorbed antisera decreased gradually according to the repetition of adsorptions before the antisera were specified. The similar phenomena were observed in IgG, IgA and IgM antibodies. Antisera adsorbed twice, of which DCP decreased little in the course of the adsorptions, were specified by the neutralization method with M and N active substance obtained by the phenol-saline extraction. The neutralized antisera gave evidently group-specific reaction on GSDCM, and the minimum titers were usually 1:8 to 1:16, which varied according to the proportion of antisera to the substance. Therefore, it might be considered that the neutralization is a suitable method to prepare the anti-M and -N sera for GSDCM.

Electron microscopic observations of the parotid gland and the pancreas were done on rats with acute pancreatitis, obstructive jaundice, ileus or peritonitis produced as reported previously. In acute pancreatitis the ultrastructural changes noted in the parotid acinar cell were an irregular arrangement of the acinar cells associated with atrophy, decrease in number of secretory granules, dilatation of cisternae of the granular endoplasmic reticulum and appearance of lipid droplets. In other sets of experiments, however, these ultrastructural changes were not noted; the appearance of lipid droplets was the only finding in the obstructive jaundice produced by ligation of common bile duct at the hilar portion of liver. The ultrastructural features of the parotid acinar cells in the experimentally produced pancreatitis were compatible with histological findings previously reported. It is conceivable that the ultrastructural changes, except appearance of lipid droplets, in the parotid gland are induced by pancreatitis. The results of the present study provide additional evidence for the close relationship between pancreas and parotid gland.

Normal human bile was collected under aseptic condition. Subsequently, it was frozen without preservative until use. A mixture of 40 bile samples was fractionated centrifugally. in the presence of salts. The resulting fractions showed strong coagulating effects on calcium carbonate suspension in water. The major fractions contained sulfated glycoproteins as the principal component and small amounts of acidic glycosaminoglycans. The present data together with previous observations suggest that sulfated glycoproteins and acidic glycosaminoglycans detected in the present normal bile samples might be a result of bacterial infection.

Inflammatory proteases were identified in the euglobulin fraction of Arthus skin lesions in guinea pigs and were partially purified by GE-cellulose column chromatography. The protease activity was partly SH-dependent and showed a pH optimum at 7.1. Intradermal injection of this protease fraction, whether activated with cysteine or not, into normal guinea pig back skin produced a marked inflammation similar to the reaction of original Arthus lesions.

The subjects of this study were a hereditary typical acatalasemia and a hereditary atypical acatalasemia. In the latter case, the children did not show hypocatala-semia but one of the cousins proved to be hypocatalasemia. The abdominal muscle and the vermiform appendix from the case of typical acatalasemia had only a minute catalase activity. The liver from the case of atypical acatalasemia indicated a moderate catalase activity and catalase protein was observed in it.

Immunoglobulin classes of antibodies to salmon sperm deoxyribonucleic acid (ssDNA) induced by heat-denatured ssDNA-methylated bovine serum albumin (MBSA) conjugates were separated and characterized, and an anamnestic response to the conjugates was reinvestigated. (1) Antibodies to ssDNA were present in both IgM and IgG. On the contrary, antibodies to polyA•polyU and polyI•polyC occurred exclusively in IgG. (2) IgM and IgG were purified by appropriate combinations of gel filtration, ammonium sulfate fractionation, sucrose density centrifugation and DEAE-cellulose chromatography, and their antibody activities were reconfirmed. (3) A single dose of a ssDNA-MBSA complex did not induce primary antibody synthesis in rabbits, but booster injections elicited the antibody. One year after stopping the booster injection, the immunological memory for antibody production almost disappeared, and the antibody producing capacity fell remark-ably.

Two autopsy cases of adenoid cystic carcinoma of the esophagus were reported. Poor clinical course and com-plexity in tumor histology were remarked especially in Case 1. The histogenesis was discussed with reference to the counterpart of other organs.

Subtypes of hepatitis B antigen (HBAg, Australia antigen) were investigated in 29 HBAg positive sera of asymptomatic carriers and hepatitis patients, living in different areas of Peru. Ethnically, 10 of them were from Indians and 19 from Mestizos. All of these HBAg sera were found to be subtype adw.

In an attempt to galvanically separate X- and Y-bearing sperm in human semen while maintaining it in as much a natural state as possible, we performed a series of experiments and have identified D. C., 2.3 V, 200μA, 30 minutes as the most adequate means of achieving this, with negligible interference with sperm motility, the balance of seminal electrolytes or pH. The seminal galvanization processed in this condition induced X-bearing sperm to the cathode and Y-bearing sperm to the anode, suggesting the possibility that this separately attracted semen could be used for the artificial insemination with the purpose of obtaining desired sexes in offspring. It is advised, however, that any practice with this method be limited to the field of livestock rearing.

Vaginoplasty and anoplasty were undertaken in a case (aged 8 years) of imperforate anus and perineal fistula with absence of the vagina by making use of the fistula and rectum and by the pull-through procedure, respectively. The surgical procedures employed are considered to be most appropriate for correction of these deformities. Three years later, however, the patient developed hematometra and hematosalpinx due to a functional uterus present. She underwent hysterectomy combined with left salpingo-oophorectomy. If normal uterus is noted to be present upon laparotomy, it seems important to establish spatial communication between the stump of the vagina constructed and the corpus uteri, to prevent future development of hematometra.

Heat sensitivity of tumor cells using mouse L-929 strain was examined in comparison with that of cells of this strain trans-formed by addition of DBcAMP and normal fibroblasts of adult mouse C3H strain. It was concluded that L-929 mouse tumor cells were more sensitive to heat than normal fibroblasts of C3H adult mouse and that transformed L-929 cells were more resistant to heat than L-929 cells and the former and normal fibroblasts are on the same level or the former is more resistant than the latter in their heat resistance.

In vivo study on the conjugation of 4-[14C]cortisol in the cerebrospinal fluid (CSF) of normal and CCl4-treated dogs was carried out. Ten min after the injection of 4-[14C]cortisol, about 3 ml of CSF was withdrawn and conjugation of 4-[14C]cortisol was assayed. Both sulphate and glucuronide conjugations of cortisol in the CSF of the dog treated with CCl4 in vivo were found to be decreased as compared with those in the normal dog, indicating that conjugation of cortisol in the CSF is markedly influenced by liver dysfunction.
