    KIMURA, I., OHTOMO, H. and KOGURE, K. Motor Disabilities and Effects of Thyrotropin-Releasing Hormone on Ataxia of Spinocerebellar Degeneration. Tohoku J. exp. Med., 1983, 139 (4), 331-337 - Fine aiming task was studied quantitatively in 20 patients with spinocerebellar degeneration (SCD) and 8 normal control subjects, utilizing a manual control system with random-step input signal. The transfer function of the system was an integral function and ability to perform a 'differentiating action' compensating for this integral function was analyzed. This 'differentiating action' was defective in all patients with SCD and was normal in all control subjects. A display of the dynamic characteristics of subjects on the plane defined by principal component analysis of the parameters showed definite delineation of the characteristics of patients from those of the controls. The intramuscular injection of thyrotropin-releasing hormone (TRH) resulted in an increase in amplitude of manipulation, a shortening of reaction time and consequently an improvement in motor disabilities in 65% of 20 patients with SCD.

Correlation of the RCA Activity and Acid-Base Balance in the in Vitro and in Vivo Experiments. Tohoku J. exp. Med., 1983, 139 (4) 339-348 - Changes in RCA activity are known to affect the acid-base balance, but a clear relationship between RCA activity and acid-base balance has not yet been established. The relationship between RCA activity and acid-base balance was studied in tonometry with 4 mixed gases for 6-9hr, in healthy adults at rest, in halothane anesthesia and in hypothermia. RCA activity increases when pH and HCO3-/PCO2 ratio increase and PCO2 decreases in vitro. It also increases when pH and HCO3-/PCO2 ratio decrease and the range of PCO2 broadens in vivo. The relationship between RCA activity and the HCO3-/PCO2 ratio is a fan shape, because a change of the HCO3-/ PCO2 ratio precedes the change of RCA activity. Some factor which increases the RCA activity may be produced secondarily in the body under the influence of high PCO2. Probably the acid-base balance is corrected by reduction of the RCA activity in the case of alkalosis, and the acid-base balance may be corrected by an increase in RCA activity in the case of acidosis

    KOYAMA K., ITO, K., ASANUMA, Y. and SATO, T. Hepatic Venography in Portal Hypertension by Balloon Catheter. Tohoku J. exp. Med., 1983, 139 (4), 349-354 - The hepatic venography and wedged hepatic venous pressure measurement were performed in 35 patients with portal hypertension using a balloon catheter, and the results were compared with those obtained by the conventional method. It was revealed that the balloon catheter visualized a broader area of the liver and showed more accurate wedged hepatic venous pressure than the conventional catheter, thereby demonstrating that this is an excellent diagnostic tool for patients with portal hypertension.

    SAITO, N., SUZUKI, F. and ISHIDA, N. Antiviral Effect of Interferon on Influenza Virus Infection in Mice. Tohoku J. exp. Med., 1983, 139 (4), 355-363 The antiviral effect of highly purified mouse interferon (IFN) was studied on influenza virus infection in mice, by changing the routes of administration. By repeated intravenous (i.v.) administrations of IFN, no antiviral effect was observed. However, IFN was effective when the infected mice received either intraperitoneal (i.p.) or intranasal (i.n.) administrations. While all of the untreated mice died within 10 days after infection, 20% of the i.p. treated mice and 67% of the i.n. treated mice survived more than 30 days. To explain these different effects after administration by different routes, the distribution of given IFN in mouse organs was examined. Fifteen min after i.v. injection, only a low-titered IFN was detected in the serum, lung, spleen and liver of the mice and soon disappeared. However, a relatively high-titered IFN was detected 30min after i.p, administration in the lung and serum. After i.n. administration, high-titered IFN was detected in the lung within 15min after administration and continued for 90min. Thus the findings demonstrated that the maintenance of a detectable amount of IFN in mice organs is dependent on the route of administration, and the higher the titer of IFN in the lung, the higher the protective effect of IFN against influenza virus infection.

Abnormalities of Lipoprotein and Carbohydrate Metabolism in Degenerative Diseases of the Nervous System - Motor Neuron Disease and Spinocerebellar Degeneration. Tohoku J. exp. Med., 1983, 139 (4), 365-376-The levels of plasma high density lipoprotein (HDL) cholesterol and plasma triglyceride were determined in 44 patients with motor neuron disease (MND) and in 36 patients with spinocerebellar degeneration (SCD). In both groups the HDL cholesterol levels were significantly lower than those in healthy controls, whereas the plasma triglyceride levels were higher than those in the controls. Glucose levels during the oral glucose tolerance test (GTT) were significantly higher in both MND and SCD groups than in healthy controls. Immunoreactive insulin (IRI) levels during GTT were rather higher at 0 and 120min in both MND and SCD groups than in healthy controls. Glycosylated hemoglobin levels were lower in MND than in the controls. These results indicate that both lipoprotein and carbohydrate metabolisms were impaired in MND and SCD groups. The possibility was presented that denervation might play a part in the pathogenesis of abnormalities of lipoprotein and carbohydrate metabolisms.

Med., 1983, 139 (4), 377-387 - A new epithelial cell line (TE-3) has been established from a metastatic lesion at the right chest wall which was originated from a well differentiated human squamous cell carcincma of the esophagus. TE-3 has been subcultured more than 150 times for 3 years and 1 month. Cultured cells have grown as isolated colonies of epithelial cells. The average doubling time of the TE-3 cell line was 48hr and the plating efficiency was 10 to 40% in MEM supplemented with 8% fetal calf serum. Distinctive marker chromosomes and a male karyotype were present in TE-3. Electron microscopic examination of the TE-3 cells disclosed the presence of desmosomes and microvilli in connection of the cells which were rich in cell organelles. In early passages of culture, the cytoplasm of cells was slightly positive with PAS stain but negative with Sudan III or mucicarmin stain. Heterotransplantation of the culture cells to BALB/c nude mice produced tumors, the histological appearance of which was similar to that of the original one. The carcinoembryonic antigen level of the medium in the confluent culture of TE-3 was 2ng/104 cells.

    KAN, M. and YAMANE, I. Oxygen-Dependent Growth Declining and Effect of Vitamin E for Human Diploid Fibroblasts in Serum-Free, BSA-Containing Culture. Tohoku J. exp. Med., 1983, 139 (4), 389-398 - Growth promoting activity of bovine serum albumin for human diploid fibroblasts was affected by oxygen environment. When cells (2×104 cells/35mm dish) were cultured with BSA of 5g/liter supplemented RITC 80-7 under hypoxic (7%) environment, excellent growth was maintained during the period of culture, while the growth under aerobic environment markedly declined after 3 days of culture. The culture challenged with BSA under aerobic environment generated a relatively large amount of malondialdehyde (MDA) as a product of lipid peroxidation. Vitamin E restored the declining of growth and inhibited MDA formation. Also in vitro lifespan of the cells challenged with BSA markedly decreased under aerobic environment as compared with that of the cells under hypoxic environment. Vitamin E did not restore the oxygen-dependent decreasing of in vitro lifespan. On the other hand, the cells cultured with serum supplemented medium grew and phased out similarly under both oxygen environment as well as the case with BSA supplemented RITC 80-7 under hypoxic environment. These results suggest that the extent of cell proliferation is controlled in part by lipid proxidation and serum antioxidant activity is not identical from that of vitamin E.

    ABE, H. Ultrastructural Study on the Granule-Containing Cells in the Rat Autonomic Ganglia. Tohoku J. exp. Med., 1983, 139 (4), 399-409 - In the rat superior cervical ganglion, small granules, 100-150nm in diameter, and large granules, 80-280nm in diameter, were seen in the granule-containing (GC) cells. Most GC cells contained almost exclusively the small granules (SG type cell), while a few cells almost exclusively the large granules (LG type cell). The GC cells formed both efferent and cholinergic afferent synapses. The efferent synapses were about four times as many as the afferent ones. In the major pelvic ganglion of rats, SG type and LG type cells were present in comparable numbers. Cells with both types of granules were also noticed. Ovoid-shaped granules, 60×140nm with a core of moderate electron density, were observed in some SG type cells. Both afferent and efferent synapses were noted on GC cells in this ganglion. The latter were more numerous than the former. SG type cells formed more synapses than LG type cells. Based on these observations, the physiological significance of GC cells was discussed.

Tohoku J. exp. Med., 1983, 139 (4), 411-415 - A 15-month-old girl with propionic acidemia presented a failure to thrive and muscular hypotonia, who had been treated with diets low in leucine, valine, isoleucine, threonine and methionine. Supplementation with leucine gave her a catch up growth and brought about an improvement in muscular hypotonia in parallel with restoration of plasma leucine which had been below normal range during the treatment with the above diets. Oral loading test of leucine produced no ketoacidosis. These findings indicate that the restriction of leucine is not necessary in the dietary management of propionic acidemia.

Tohoku J. exp. Med., 1983, 139 (4), 417-418 - An effective method for removing leukocytes and platelets from red blood cell concentrates (RBC) reconstituted with phosphate buffer solution was investigated in vitro. Reconstituted RBC were warmed at 37°C for 1hr. Neither leukocyte count nor platelet count was decreased by the treatment. When compared with whole blood, leukocytes present in the fractions below the buffy coat layer were significantly less than those of whole blood, while the distribution of platelets in reconstituted RBC was unchanged. It is suggested that the reduction of leukocytes in red blood cells below the buffy coat layer may contribute to the preparation of RBC with a poor leukocyte.

    UZUKA, Y. and SAITO, Y. Treatment of the Blastic Crisis of Chronic Myelogenous Leukemia with the Combination of Vindesine and Predonisolone, and Cytosine Arabinoside. Tohoku J. exp. Med., 1983, 139 (4), 419-420 -Sixteen patients with chronic myelogenous leukemia in blast crisis were classified into three subgroups on the basis of blast cell morphology and a relation to the chemotherapeutic responsiveness was investigated. All of the four patients with lymphoid morphology, and seven of the eight with myeloid morphology (88%) achieved complete remission while none of the four with monocytoid morphology achieved complete remission.

    SAITO, Y. and UZUKA, Y. Treatment of Acute Lymphoblastic Leukemia in Adults with Combination Chemotherapy Incorporating Vindesine and Prednisolone. Tohoku J. exp. Med., 1983, 139 (4), 421-422 - Seven adult patients with acute lymphoblastic leukemia (ALL) were treated with induction regimen incorporating vindesine (VDS) and prednisolone (VDS-P). After achieving complete remission, multidrug regimens for remission consolidation and maintenance were given. All patients achieved complete remission including five cases of continuons complete remission.

Med., 1983, 139 (4), 423-424 - Eight distinct polypeptides with molecular weights of 130, 000 (130K, VP1), 96K (VP2), 84K (VP3), 80K (VP4), 56K (VP5), 40K (VP6), 35K (VP7) and 28K (VP8) were identified in human rotavirus (Wa strain) particles. Only VP7 was glycoprotein possessing high-mannose type oligosaccharide chain(s). Tunicamycin markedly suppressed the incorporation of [3H]glucosaminein to the VP7. This glycosylation pattern is very similar to that reported in simian rotavirus SA11.
