    HOSOKAWA, M., MASU, S. and SEIJI, M. Immunofluorescence Studies on Civatte Bodies and Dyskeratotic Cells with Anti-Keratin Antibody. Tohoku J. exp. Med., 1981, 135 (3), 219-229-So-called Civatte bodies and dyskeratotic cells were investigated in some skin disorders by using indirect immunofluorescence techniques with anti-human keratin antibody. In the disorders with lichenoid tissue reaction such as lichen planus and DLE, Civatte bodies were observed in the lower epidermis and upper dermis and they reacted distinctly to the anti-keratin antibody. In malignant skin tumors which show dyskeratotic cells in the epidermis, such as basal cell epithelioma and Bowen's disease, dyskeratotic cells were more clearly reacted to the antibody than other keratinocytes. These observations present additional new evidence for the hypothesis that Civatte bodies are derived from tonofilaments of keratinocytes.

Med., 1981, 135 (3), 231-236 A total of 191 patients with hemispheric lesion mainly due to CVD were examined by the finger-tapping test, and their disturbances of rhythm formation, `hastening phenomenon' (HP), were compared with those of Parkinson's disease. HP was observed in 56% of the right and 49% of the left hemiplegic patients. HP in the intact hand of the patient was highly correlated with that of the affected hand. CT scans were examined in 86 of 191 patients, and HP was specifically correlated with unilateral striatal lesions. Thus, the disturbances of rhythm formation in hemiplegic, together with those in Parkinson's disease, Huntington's chorea, SND and OPCA, were attributed either to organic or to functional deficits in the striatum.

Med., 1981, 135 (3), 237-245 This study was undertaken to find out some clue to detect early relapse of adult acute leukemia which had been in remission. Laboratory data such as LDH, erythrocyte sedimentation, or immunoglobulin level did not show any difference at the time of relapse from the remission period. Complete blood count with reticulocyte count was evaluated every two weeks retrospectively for 12 weeks before relapse. No significant change was observed at the time of relapse. However, both the percentage and absolute number of peripheral lymphocytes significantly increased, whereas percentages of both T and B cells markedly decreased at the time of relapse, suggesting some derangement of lymphocyte function. Results in bone marrow culture did not yield any remarkable difference. It was suggested from this study that it is the time of prerelapse of adult acute leukemia when lymphocytes show an increase in either percentage or absolute number in the patients who are followed under their maintenance therapy schedule.

Med., 1981, 135 (3), 247-253-The effect of RES blockade on the survival of heat-damaged and antibody coated red blood cells was evaluated in C3H mice. RES function of C3H mice was determined by the survival of 59Fe-labeled, heat-damaged red blood cells at 49°C for 30min. When animals were treated with an intravenous injection of non-labeled, heat-damaged red blood cells for the blockade of RES, red blood cell destruction was depressed in non-splenectomized and splenectomized mice. In the survival or organ distribution of 59Fe-labeled, antibody-coated red blood cells, no difference was noted between animals with and without blockade. These findings suggest that heat-damaged red blood cells were destructed predominantly in the splenic RES and antibody coated cells were not broken down only by the splenic RES.

Med., 1981, 135 (3), 255-257-Serum FSH, prolactin and HCG response patterns to 4-hr infusion of LHRH were investigated in six women in the first trimester of pregnancy. FSH did not show response until 60min after beginning of the infusion and then it increased gradually. Serum prolactin and HCG levels were almost constant during the LHRH infusion.

    KITAMURA, M., SUDA, M., NISHIHIRA, T., WATANABE, T. and KASAI, M. Heterotransplantation of Human Esophageal Carcinoma to Nude Mice. Tohoku J. exp. Med., 1981, 135 (3), 259-264 - Transplantation to nude mice of 38 tumors originating from 36 human esophageal carcinomas was attempted. Growth of the grafts was obtained in 17 cases and 3 to 11 serial passages were performed in 4 cases. Tumors inoculated into subcutaneous tissue grew locally, but no metastasis nor local invasions were observed. The histological appearances of the grafts were similar to original tumors.

Med., 1981, 135 (3), 265-273-Rat lung phosphatidylglycerol contained about 21% of saturated species, a predominant of which was dipalmitoyl. The biosynthetic features of molecular species of the phospholipid were investigated by incubating rat lung slices with radioactive precursors. The labeling profiles of molecular species of phosphatidyl-glycerol by [2-3H]glycerol and [1-14C]palmitate were significantly different from that of diacylglycerol and phosphatidylcholine. Mono- and dienoic species of phosphatidylglycerol were shown to have extremely high reactivity with [3H]glycerol, but the labeling of saturated species were almost half of the saturated diacylglycerol. The rate of [14C]palmitate incorporation was significantly lower in the saturated species of phosphatidylglycerol as compared to that in diacylglycerol and phosphatidylcholine. It was also noted that the lung tissue can utilize 1-[1-14C] palmitoyl lysophosphatidylglycerol to form tetraenoic and polyenoic phosphatidylglycerol by the direct acylation and saturated phosphatidylglycerol probably by the transacylation. The data indicate that the saturated species of phosphatidylglycerol may be synthesized de novo via phosphatidic acid CDP-diacylglycerol route and also via transacylation pathway, but it may not be synthesized by the direct acylation pathway.

Med., 1981, 135 (3), 275-279-A particulate fraction was separated from endometrial scrapings of uterus of hormone-treated non-ovariectomized or intact rabbit. The effects of ovarial hormones on the incorporation of [35S]sulfate from 3'-[35S]phosphoadenosine 5'- phosphosulfate (PAPS) into endogenous acceptors in the particulate fractions were studied. Treatment of the animal with estrogen elevated the incorporation of [35S]sulfate, but exogenous progesterone suppressed this effect. The results of DEAE-Sephadex A-25 (Cl- form) column chromatography of the pronase digest of the 35S-labeled substances indicated that all the subfractions were sensitive to the hormones. The present observations, together with previous findings, suggest that the sulfation of sulfated glycoconjugates in the particulate fractions is stimulated by exogenous estrogen, but exogenous progesterone suppressed this effect, and also that endogenous ovarial hormones exert little influence on the effects of the present doses of these exogenous hormones on the sulfation. Therefore, non-ovariectomized rabbit can be used for rough evaluation of the effects of regular doses of these exogenous hormones on the sulfation of sulfated glycoconjugates in the uterine endometrium.

Correlation between the Level of Urinary D-Glucaric Acid and the Degree of Activation of Masked Compound (FT-207) in Cancer Patients. Tohoku J. exp. Med., 1981, 135 (3), 281-290-Phenobarbital stimulates the induction of liver microsomal drug-metabolizing enzyme, namely, cytochrome P-450, which enhances the rate of conversion of FT-207 to 5-FU, the active substance. When FT-207 is administered in combination with phenobarbital to cancer patients, the fluctuation in level of the drug-metabolizing enzyme, cytochrome P-450, should be taken into consideration. Therefore, it was investigated whether the urinary level of D-glucaric acid could be of value as an indicator for the evaluation of the activation of masked compounds, such as FT-207. The level of D-glucaric acid in urine was lower in cancer patients than in normal controls. The correlation between the level of urinary D-glucaric acid and that of 5-FU, which is an active metabolite of FT-207, in blood was statistically significant. The level of D-glucaric acid in urine was of use as an indicator for the evaluation of the activation of masked compounds, such as FT-207, in cancer chemotherapy.

Development of Malignant Hypertension in Patients with Uremia under Hemodialysis: A Case Report and Discussions on Its Etiology. Tohoku J. exp. Med., 1981, 135 (3), 291-299 A woman, now 28 years old, was diagnosed 6 years ago as chronic glomerulonephritis by renal biopsy. From August 15, 1975 she complained of nausea, loss of appetite and weight (about 7kg within 2 weeks). Severe hypertension (200/130mm/g), hyponatremia (123 mEq/liter), anemia, elevated plasma renin activity (PRA), advanced azotemia, and eye ground changes of KW-II were found. Dialysis treatment was started on September 2, 1975. From November 1975 massive amounts of sodium (5, 000 mEq or more monthly) and water (26 liters or more monthly) were removed by the dialysis. These intensive dialyses resulted in an elevated PRA with recurrence of severe hypertension. At the end of March 1976 she became almost blind with retinopathy of KW-IV. Potent hypotensive drugs including β-blockers were administered, but no improvements were obtained. On March 31, 1976 nephrectomy was performed to save her life. Marked hyalinization of glomeruli and heavy thickening of intima in interlobular arteries were found in the removed kidneys. Renal artery stenosis was not recognized either macroscopically or histologically. In this patient, the amount of sodium removed by the dialysis was dependent on her diastolic blood pressure and sodium concentration of the dialysate. It may be concluded that too enthusiastic dialysis may develop malignant hypertension due to excessive renin release.

Med., 1981, 135 (3), 301-307 -The urinary bladder is assumed to be an incompressible isotropic material like a rubber. Then, by making use of the finite deformation theory for such a hyperelastic continuum, we have calculated the intravesical pressure and the mural stress of the bladder as a function of its volume. The formula for the mural stress contains two parameters. These parameters are determined by the uniaxial extension test of the dog bladder sample. The calculation result shows that the pressure is relatively flat while the stress continues to rise with the volume. This is the typical behavior in the normal living cystometrogram. From these analysis, therefore, we conclude that such a behavior comes from the elasticity of the bladder itself, and requires no explanation in terms of reflex inhibitory relaxation.

Endoscopic Gastric Polypectomy Using High Frequency Current: Its Significance for Total Biopsy of Gastric Polyp. Tohoku J. exp. Med., 1981, 135 (3), 309-322-Endoscopic polypectomy using high frequency current was performed for 281 out of 345 gastric polyps (230 cases), while the remaining 64 small polyps were cauterized. Histopathologic examinations carried out in 274 out of the 281 polypectomized polyps confirmed gastritis polyposa in 41 lesions, hyperplastic polyps in 216 including 3 cases of neoplasm, atypical epithelium in 4, submucosal tumor in 2, and polypoid cancer in 1. This procedure of endoscopic polypectomy has proved to enable histologic examination of a polyp as a whole, and to allow a highly risk-free endoscopic treatment of polyps. In the currently observed 216 hyperplastic polyps, 3(1.4%) were found to have neoplastic changes.

Abnormalities of Iodine and Protein in the Thyroid of Patients with Hashimoto's Thyroiditis.Tohoku J. exp. Med., 1981, 135 (3), 323-330-Iodine content, protein content and percentage of thyroglobulin to total protein in the thyroid glands were studied in two groups of 10 patients with Hashimoto's thyroiditis (5 euthyroid and 5 hypothyroid patients) and in 6 normal subject. Iodine content was significantly decreased in both hypothyroid (24+29μg/g wet weight of thyroid tissue, mean±S.D., p<0.01) and euthyroid (96+52, p<0.01) groups of Hashimoto's thyroiditis compared with those in controls (779+37). The value of hypothyroid group was significantly smaller than that of euthyroid group (p<0.05). A significant negative correlation was obtained between thyroidal iodine content and serum TSH (r=-0.68, y=-0.41x+55.6, p<0.05). Thyroidal protein and percentage of thyroglobulin to total thyroidal protein were also decreased in Hashimoto's thyroiditis compared with controls, while there was no difference between euthyroid and hypothyroid groups in these values. The percentage of thyroglobulin-bound 131I obtained by in vitro iodination of thyroglobulin in Hashimoto's thyroiditis appeared to be decreased in 2 cases of hypothyroid group. Some immunological reaction occurring in Hashimoto's thyroiditis may change the structure of thyroglobulin and in turn the normal iodination of thyroglobulin may be disturbed.-Hashimoto's thyroiditis; thyroidal iodine; thyroidal protein; thyroglobulin; in vitro iodination

Med., 1981, 135 (3), 331-332-A 44-year-old woman with atypical gyrate atrophy and iminoglycinuria was described. The serum ornithine level and ornithine-ketoacid transaminase (OKT) activity were both normal. Urinary excretion of proline, hydroxyproline and glycine was markedly increased. This finding, together with the existence of gyrate atrophy with hyperornithinemia due to OKT deficiency, suggests that proline deficiency in the chorioretinal tissues may concern the development of gyrate atrophy.

Effect of Pyridoxal 5-Phosphate on the Activity of GOT Isozyme in Plasma from Patients with Duchenne Muscular Dystrophy. Tohoku J. exp. Med., 1981, 135 (3), 333-334-GOT isozyme (soluble GOT, s-GOT and mitochondrial GOT, m-GOT) activities with and without reactivation by the co-enzyme pyridoxal 5-phosphate (PALP) in plasma from 42 patients with Duchenne muscular dystrophy (DMD), and 11 normal controls were investigated. The magnitude of the enzyme activities in patients at the early stage of DMD greatly exceeded those of controls. As the disease progressed, the levels gradually declined, but late stage values were still higher than the control values except s-GOT with PALP. At the mid stage as well as the late stage, the increase in m-GOT activity was approximately to double the unstimulated activity, indicating that, in these patients, as much as half the m-GOT in the circulation was in the inactive apoenzyme form, whereas s-GOT showed negligible increase following PALP addition at any stage.
