Med., 1985, 147 (3), 221-234 - Alteration in lipid metabolism of Ascaris lumbricoides eggs during development in either air or nitrogen gas was studied using acetate-1-14C and labeled fatty acids. It is thought that formation of 14C-lipids and 14C-palmitoleate from acetate-1-14C and palmitate-1-14C in developing eggs may be influenced by the concentration of molecular oxygen in the medium. Acetate-1-14C was incorporated into palmitate, palmitoleate and unsaturated 18-carbon acid of fatty acids in the eggs, but the incorporation into saturated 18-carbon acid was slight. Radioactive fatty acid methyl esters were majer lipid component of the radioactive lipid classes in the first and second stage larvae incubated with palmitate-1-14C. Isopentadecanoic acid, which is a small component of fatty acids in Ascaris eggs, contains a high percentage of the total radioactivity in the second stage larvae incubated with acetate-1-14C under both air and nitrogen. It appears that a conversion of the fatty acid biosynthesis system may occur during development in to the second stage larvae.

Diagnosis of Submucosal Tumors by Injecting a Water Soluble Contrast Medium: Diagnosis of Extra-Gastric Tumors and Gastric Varices Tohoku J. exp. Med., 147 (3), 235-245 - Extra-gastric compression caused tumescent lesions that are difficult to differentiate from gastric submucosal tumors, and gastric varices similar in appearance to circumscribed tumors were sometimes experienced clinically. Up to now, the differential diagnosis of these lesions has been done by the palpation through x-ray examination or the tactile test under endoscopic examination. Even by the recent use of the CT scan, the differential diagnosis still remains unsatisfactory except in a few specific cases. Under these present circumstances, submucosography is recommended for routine screening test for outpatients. Our method is simple, safe and time-saving. Recently, it has become easy to diagnose hemangiomas as well as varices in the stomach by application of submucosography. Accordingly, in the cases of vascular tumors or tumescent lesions caused by extra-gastric compression, the risk of incidental major bleeding or perforation can be prevented by using the submucosography.

Med., 1985, 147 (3) 247-259 - Dogs with a vagally denervated (Heidenhain) pouch and a gastric fistula were used to investigate the humoral mechanism which would affect the gastric acid secretion following acute intragastric ethanol administration. Ethanol solutions induced a dose-related secretion of gastric acid. Although plasma gastrin levels increased after the loading of both 20% and 30% ethanol solutions, there was a discrepancy between total acid output and the integrated gastrin response. Plasma secretin levels also augmented after the administration of ethanol solutions, with a delay of about one hour after the onset of acid secretion. The response of gastric inhibitory polypeptide (GIP) to ethanol was very slight, similar to that of insulin. There was a significant rise in plasma glucose levels after the instillation of 30% ethanol solution as in the case of liquid meal. It is concluded that gastrin may be merely one of the factors which stimulate acid secretion after ethanol administration, and that gastric acid would have a close relationship with secretin release. It is also probable that acid is not an effecitve stimulant to the release of GIP.

Tohoku J. exp. Med., 1985, 147 (3), 261-265 - The distributional changes of type IV collagen and laminin with normal maturation of human placentae were examined in relation to those of fibronectin by the histochemical methods including immunofluorescence staining. In the early chorionic villi, these components were detected along the trophoblastic basement membrane, around the fetal blood vessels, and in the villous stroma. Laminin was detected also in the pericellular matrices of nonvillous cytotrophoblasts where type IV collagen was rarely detected. In the late and term placentae, laminin and type IV collagen were detected along the trophoblastic basement membrane, while this structure was virtually not stained for fibronectin. These observations suggest that type IV collagen and laminin are the constituents of the trophoblastic basement membrane throughout the maturation period of the placentae, while fibronectin is a transient constituent.

    TAKEDA, K. and HIRAIA, K. ABO Blood Group Antigens of Human Spermatozoa. Tohoku J. exp. Med., 1985, 147 (3), 267-272 - ABO blood group antigens on the membrane of human spermatozoa were investigated with the peroxidase-labelled antibody test. Spermatozoa from O secretor were incubated with saliva or seminal plasma of A and B secretors, but the specific peroxidase staining was negative. This result indicates that blood group antigens on human spermatozoa originate from spermatozoon itself, but not from seminal plasma.

Med., 1985, 147 (3) 273-280 - We studied here the clinical significance of GHRF test in evaluating therapeutic results in patients with acromegaly. Three cases of acromegaly were studied at pre- and posttreatment. Case 1 is a 42 year old woman, and her plasma GH showed slight increases responding to TRH and arginine administration before therapy. Eleven years after the pituitary irradiation, basal plasma GH decreased to low normal ranges and showed no response to the above two agents. Only an insufficient increase in plasma GH was observed after GHRF injection. This case would have pituitary lesions not only in the tumor somatotrophs but also in the normal somatotrophs. Case 2 is a 38 year old man. His plasma GH showed a marked increase after TRH injection. To GHRF administration, however, no clear response was observed. After transsphenoidal adenomectomy, the aberrant responses to TRH was still observed despite the marked GH decrease to the near normal ranges. Interestingly, plasma GH became responsive to GHRF after the operation. It seems that the adenomectomy was inadequate in this case and the normal somatotrophs recovered the responsiveness to GHRF. Case 3 is a 53 year old woman, and she showed aberrant GH responses to TRH and to L-dopa. Her plasma GH clearly increased after GHRF injection. Postoperatively, the abn ormalresponses of plasma GH disappeared completely, while a normal GH response was observed following GHRF injection. It is suggested that the tumor somatotrophs were removed completely, and the normal somatotrophs were selectively preserved in this case. These findings indicate that GHRF test is clinically useful to evaluate therapeutic results in patients with acromegaly.

Med., 1985, 147 -(3), 281-293 - The enzymatic activity of bacterial β-glucuronidase plays an essential role in the formation of calcium bilirubinate in bile. There are, however, many unsettled problems such as methodology of the assay for its enzymatic activity. In the present study (1) the azopigments from mono-conjugated bilirubin (MCB) and unconjugated bilirubin (UCB) in native bile were semiquantitatively determined, (2) the deconjugation of conjugated bilirubin (CB) in bile was estimated with azopigment analysis and (3) factors affecting the deconjugation of CB in bile, especially for pH value, were investigated. CB in bile was stable at physiologic pH during 6-hr incubation at 37°C, but was hydrolyzed at alkaline pH. At physiologic pH, addition of β-glucuronidase from E. coli hydrolyzed CB in bile and increased MCB and UCB in bile. Based upon the results mentioned above, it is suggested that alkaline pH and enzymatic activity of β-glucuronidase should cause the increase of UCB in bile. It can be said that β-glucuronidase is essential for the formation of calcium bilirubinate gallstone at physiologic pH.

Med., 1985, 147 (3), 295-300 - Heparan sulfate was prepared from the pronase digest of the porcine kidney and modified chemically to yield N-desulfated, N-, O-desulfated, and N-O-desulfated N-acetylated heparan sulfates. Heparan sulfate and its modifications thus obtained were used as substrates to measure the sulfotransferase activity in the microsomal fraction of the endometrium of the rabbit uterus. N-, O-Desulfated heparan sulfate was the best acceptor of sulfate from 3'-phosphoadenylyl [35S] sulfate and intact heparan sulfate was the poorest one. The products of enzymatic reaction were treated with nitrous acid and examined by gel-filtration and electrophoresis. The results indicated that sulfate was almost exclusively incorporated as N-sulfate into all the present substrates.

    TAKAHASHI, Y., TAKAHASHI, T., TAKAHASHI, W. and SATO, T. Morphometrical Evaluation of Extrahepatic Bile Ducts in Reference to Their Structural Changes with Aging. Tohoku J. exp. Med., 1985, 147 (3), 301-309 - Forty-three autopsy cases with neither history nor anatomic evidence of hepatobiliary or pancreatic diseases were submitted to morphometrical evaluation of the internal diameter of the common hepatic duct. At a cross section of the duct the perimeter length of the mucosal basement membrane was measured to calculate its diameter D to be obtained when it was stretched into a circle. The value of D was found to correlate significantly with age, with the upper rejection limit (5% level) growingly elevated from 4.8mm at 20 years to 7.5mm at 80 years. This implies that the age of patient should be taken into account in judging clinically whether or not bile ducts actually are dilated. The density in the ductal wall of elastic fibers was also determined and was shown to increase as the senile dilatation progressed, a sign of adaptive reinforcement of wall tissue that is liable to gradual hysteresis under chronic tension load.

Dobutamine administered intra-arterially at the bifurcation of the iliac artery produced a rise in pressure and shortening along the transverse axis in the urethra.

    HASHIMOTO, L., OUCHI, K., KOYAMA, K. and SATO, T. Superselective Arterial Embolization of the Rat Liver. Tohoku J. exp. Med., 1985, 147 (3), 315-316 -The superselective transcatheter arterial embolization has currently become an accepted treatment of hepatic tumors. We have developed an experimental model for the evaluation of this therapy in the rat liver. Using the retrograde catheterization of the superior pancreaticoduodenal artery, only the peripheral intrahepatic arterial branches and sinusoids of the median and left lobes of the liver were embolized with 1.2 and 1.5mg of Gelfoam powder. This method resembles the clinical practice because the superselective embolization could be performed adequately and allows the comparative evaluation of the responses of the embolized and non-embolized lobes.
