    YOSHIDA, I. Early Interferon Production in Human Diploid Cells Induced by Newcastle Disease Virus in the Presence of Protein Synthesis Inhibitors. Tohoku J. exp. Med., 1985, 147(2), 113-124-When human diploid cells were induced by Newcastle disease virus (NDV) in the presence of cycloheximide or puromycin, an early interferon was produced up to 4hr after induction, but was not produced without these reagents. Early interferon production in rabbit kidney cells induced by NDV in the presence of cycloheximide was not observed. This early interferon production was also induced in response to hydroxylamine-treated NDV, ultraviolet light-irradiated NDV and Sepharose-coupled NDV, which had no or low induciblity of late interferon, and was inhibited by pretreatment with actinomycin D. Protein and RNA synthesis were enhanced in the cells which were treated with cycloheximide and then washed out as in the induction process. From these findings it has been suggested that the induction of early interferon synthesis in the cells pulse-treated with protein synthesis inhibitors, was triggered by an interaction between viral envelope and cell membrane.

Imbalance of T Cell Subsets in Cancer Patients and Its Modification with Bestatin, a Small Molecular Immunomodifier. Tohoku J. exp. Med., 1985, 147 (2), 125-133-The peripheral T lymphocyte subsets and natural killer (NK) cells of patients with cancer were studied by monoclonal antibody assays in comparison with those of non-cancer individuals. An imbalance of immunoregulatory T cell subsets was frequently found in the group of cancer patients (n=26), being characterized by a relatively high proportion of OKT8+ cells (45.0± 12.0%, p<0.01) and a low OKT4+/OKT8+ ratio (1.29±0.56, p<0.05). T cell level showed no significant difference as assessed by either OKT3 or rosette formation with sheep erythrocytes (E). The effect of bestatin was examined in 10 of these cancer patients. The relative proportion of T cell subsets tended to normalize 2 to 6 weeks after starting the daily doses of 30mg per body. The mean percentage of OKT8+ cells decreased from 47.2 to 33.4 (p<0.01), while the helper to suppressor ratio increased from 1.18 to 1.79 (p<0.01). A significant increase was also found in the percentage and number of OKT4+ cells and HNK-1+ cells after the drug administration, while T cell level tended to converge to a normal range.

Determination by Enzyme-Linked Immunosorbent Assay (ELISA) of Specific IgG Antibody Activities for Diagnosis of Farmer's Lung Disease. Tohoku J. exp. Med., 1985, 147 (2), 135-144-The specific IgG antibodies to thermophilic actinomycetes in the sera from patients with farmer's lung disease were quantitated by an ELISA and the results thereof were compared with those obtained by the standard double immunodiffusion assay (Ouchterlony's method). All sera from patients with farmer's lung disease were precipitin positive to Thermoactinomyces vulgaris and/or Micropolyspora faeni by the Ouchterlony's method, and revealed significantly high levels of IgG antibody activities against these antigens by ELISA. We have found 18 precipitin-positive but asymptomatic subjects in a survey of the dairy farming population of the northern area of IWATE Prefecture. Their sera were precipitin positive to antigens related to the farmer's lung disease, however IgG antibody activities as determined by ELISA to these antigens was significantly lower in the group of asymptomatics than in the group of symptomatics. Precipitin negative dairy farming controls and normal controls living in the urban area revealed low antibody activities as determined by ELISA. From these studies, we concluded that the assay of specific IgG antibody activity to thermophilic actinomyces by ELISA is useful for the diagnosis and screening of farmer's lung disease.

Purification and Characterization of the Specific Binding Protein for Platelet Activating Factor (1-O-Alkyl-2-Acetyl-sn-Glycero-3-Phosphocholine) from Human Platelets. Tohoku J. exp. Med., 1985, 147 (2), 145-152 The binding protein to platelet activating factor (PAF) was solubilized with 2% Triton X-100 from human platelet membranes and purified approximately 23-fold by sequential chromatography on DEAE-cellulose, CM-cellulose and Sephadex G-200. The final preparation migrated as a single protein band correspoding to a radioactive peak of [3H] PAF bound on polyacrylamide disc gel electrophoresis. The molecular weight of the native form of the binding protein was estimated to be approximately 160, 000 daltons by Sephadex G-200 column chromatography, and its isoelectric point was near 8.0. Exposure of the platelet membranes to an excess of unlabeled PAF diminished the binding of [3H] PAF.

Med., 1985, 147 (2), 153-155-Isoproterenol produced a concentration-dependent increase in contractile force of rabbit upper renal pelvic smooth muscle. Conversely, isoproterenol produced a concentration-dependent decrease in contractile force of rabbit lower renal pelvic smooth muscle. These data show that in response to isoproterenol upper renal pelvic smooth muscle, which is thought to act as a pacemaker region for pelviureteral peristalsis, resembles cardic muscles, whereas lower pelvic smooth muscle is similar to ureter and other smooth muscles.

A Method for Making Chronic Fetal Preparation in the Lamb: Results of 4 Years' Experiences. Tohoku J. exp. Med., 1985, 147 (2), 157-168-During the period between 1980 and 1983, a versatile experimental model for fetal physiology (chronic fetal preparation) was made using 26 pregnant Corriedale and Suffolk sheep. The sheep fetuses were removed from the uterus at the gestational ages of from 96 to 133 days. After performing cannulation of the carotid arteries, the jugular veins, and the trachea of the fetuses, applying tripolar cardioelectrodes, and attaching an occluder of the umbilical cord, the fetuses were retuned to the uterus to complete the gestational period. The time left until the termination of the postoperative pregnancy and available for various intrauterine monitoring procedures varied from 3 to 800hr. Eleven sheep (42.3%) continued their postoperative intrauterine term for 5 days or more. The continuation of the postoperative gestation for longer than 5 days indicated a return to the physiological state on the evidence of the measured PO2, PCO2 and pH values and rapid irregular breathing movements.

Med., 1985, 147(2), 169-176-The following conclusions have been drawn from electron-microscopic three-dimentional observations on the intermediate filaments (IFs) of the Squamous epithelium from the uterine cervix. (1) IFs form a three-dimensional reticular network and wrap around the intracellular periphery of cells extending from the basal to the superficial layers. (2) In basal and parabasal cells, the IFs are distributed in a pattern radiating from the perinuclear location toward the cellular periphery. (3) The use of PEG-embedding method after detergent-extraction with Triton X-100 and saponin is suitable for the three-dimensional observations on IFs of stratified squamous epithelium from the uterine cervix.

Med., 1985, 147 (2), 177-181-Atrioventricular (AV) block was produced in animals for experimental purposes by use of a newly devised cannula. It has bipolar electrodes; one electrode on the needle tip and the other 5.0mm proximally from the tip. Otherwise, the cannula was electrically completely insulated. The cannula was inserted from the right external jugular vein and advanced close to the AV ring. While His bundle electrogram was obtained with the electrodes, the AV node was accessed. The injection site was determined as the point where His bundle electrogram can be well recorded and the atrial electrogram is close to or larger than the ventricular electrogram in amplitude. Xylocain solution, 0.2 to 0.4ml, was injected with the syringe attached on the other end of the cannula. Transient AV block occurred when the needle tip was properly positioned. Then, 0.2ml to 0.4ml of anhydrous alcohol was added to make permanent a AV block. A possible application of this technique was implicated.

Med., 1985, 147 (2), 183-190-In order to clarify the role of the vagus nerve in the secretion of gastric inhibitory polypeptide (GIP), experiments were performed on dogs. Response of plasma GIP to intraduodenal instillation of glucose was slightly lower in a group which received atropine, than in a group of normal dogs. The response of plasma GIP to intraduodenal glucose load was not different between vagotomized dogs and normal dogs. Electric stimulation of the vagus nerve did not produce any significant changes in plasma GIP in anesthetized dogs. In conclusion, the present study indicates that the role of the vagus nerve on GIP secretion is tiny, if any, and that the nervous influence does not overcome the effect of intraluminal administration of glucose.

Med., 1985, 147 (2), 191-197-The effect of facilitating position of the lower extremity on EEG states was examined through topographical analysis of spectral power for each major frequency band in 8 healthy male subjects. Compared to neutral position, facilitating position of the left or right lower extremity produced significant changes in spectral power, especially in terms of non-specific increase at alpha band (8-12.8Hz). These results provide further evidence of arousal shift induced by facilitating position, and suggest that EEG topography is a suitable indicator of monitoring the effect of facilitating position.

Med., 1985, 147 (2), 199-212-In vitro effects of cimetidine on the antiviral, the antiproliferative and the natural killer (NK) cell stimulating activities of human leucocyte interferon (IFN-α (Le)), and on the Sendai virus induced interferon production in lymphocytes were investigated. The antiviral activity of IFN-α (Le) was dosedependently enhanced by cimetidine, which alone did not demonstrate any antiviral action. The observed titer of the same IFN preparation titrated with 60μg/ml of cimetidine was significantly higher (p<0.05) than that without cimetidine. Cimetidine slightly but dosedependently inhibited the growth of Daudi or G-361 cells. The combination effects of these two agents were revealed to be statistically additive or synergistic. The NK cell activity of peripheral blood lymphocytes (PBL) was suppressed after a 16hr pretreatment with cimetidine. Furthermore, the interferon production in normal lymphocyte cultures was suppressed dosedependently by cimetidine. On the other hand, the augmentation of the NK cell activity by interferon was further potentiated by cimetidine.

    NAKAMURA, S. Recent Changes in Serological Markers of Hepatitis B Virus Infection in Japanese University Students. Tohoku J. exp. Med., 1985, 147 (2), 213-216-University students were screened for serological markers of hepatitis B virus infection every year. Hepatitis B surface antigen (HBsAg) determined by reversed passive hemagglutination and antibody to HBsAg (anti-HBs) by passive hemagglutination were positive in 1.7% and 7.3% of the first-year students in 1978-80, respectively, and 1.1% and 3.8% in 1981-84. The difference in the prevalences of anti-HBs between 1978-80 and 1981-84 was statistically significant (p<0.001). Hepatitis B e antigen, which was positive by less sensitive immunodiffusion in 52% of HBsAg carrier students in 1976-79, was detected by more sensitive radioimmunoassay only in 19% in 1980-84. The difference was statistically significant (p<0.01). Recently changes in hepatitis B virus infection seem to be occurring in Japanese university students.

Med., 1985, 147 (2), 217-218-Rescue of acute radiation dermatitis has already been shown feasible with cytochrome c(Sci. Rep. Res. Inst. Tohoku Univ.-C 29: 1, 1982). Interest in the probable reduction of inflammatory sequelae led to histological studies in an ultraviolet model system in the rabbit. The relative dermal thickness in the field of ultraviolet irradiation was 1.70±0.30 (5) in the untreated control and 1.31±0.07 (5) in the cytochrome c group respectively (p<0.05). Usefulness of the principle cannot be overemphasized in the strategies of radiotherapy, and in the treatment and/or prevention of actinic radiation dermatitis.

Med., 1985, 147 (2), 219-220-The culturing of normal human melanocytes from the roof of suction blisters of adult oriental volunteers was carried out. Since the epidermal roofs of blisters did not contain any fibroblasts, and since keratinocytes did not attach to the culture dishes in the presence of PMA (phorbol 12-myristate 13-acetate), many melanocytes were obtained which grew well in the presence of PMA. This method is a very simple and easy way to establish pure melanocyte cultures.
