Med., 1983, 141 (2), 125-131 The mass screening for detection of breast cancer has been conducted in Miyagi Prefecture since 1977. The main activities comprise the itinerant screening in the residential districts and the group screening of female workers at the vocational sites. The routine examination at the Cancer Detection Center is also an integral part of the activities. In the present paper, the data from the above three procedures were compared. The examinees were 49, 114 in the residential districts, 6, 725 at the vocational sites and 3, 055 at the Center. The breast cancer was detected in 51 cases (0.1%) by the itinerant residential screening, 14 (0.2%) at the vocational sites and 73 (2.4%) at the Center. The problems of the first-stage mass screening to detect breast cancer at the early stage were discussed.

Effects of Alcohol Diet from the Fetal Period on Lipid Metabolism in Rats. Tohoku J. exp. Med., 1983, 141 (2), 133-142-The effects of maternal alcohol consumption on lipid metabolism in the litter liver were examined in the rat. Rats reared from their fetal period to adulthood with liquid alcohol diet (fetal alcohol group) did not show any lipid accumulation in the livers. Liver perfusion experiments showed an increased oxygen consumption in the liver to a load of physiological concentration of palmitate. On the contrary, the response of oxygen consumption in fetal control rat livers (reared with non-alcohol liquid diet) to palmitate was found to be depressed. 14CO2 production rate from 14C-palmitate in pfused livers showed a significant increase in fetal alcohol group as compared with controls. No difference in 14C-palmitate incorporation into tissue lipid was found between fetal alcohol and fetal control groups. These data indicate that when rats are treated with alcohol from fetal period they acquire an ability to elevate FFA oxidation in the liver through metabolic adaptation.

Med., 1983, 141 (2), 143-154-To elucidate immunochemical properties of Rh-Hr antigens, influence of various solubilizers on the activity, and isolation of the antigens were investigated. As a very sensitive method to detect Rh-Hr antigens, the agglutination inhibition test was recommended in which diluted antisera (1:400-1:1, 600), papainized red cells and microtitrate plates were used. Extraction of D antigen with Brij 35 and Triton X-100 was effective, but SDS was not successful. However, D activity of SDS solubilized stroma was recovered by adding reductants. The activity was slightly demonstrated at the preparation from D negative red cells. The influence of the reductants was discussed. SDS solubilized stroma was fractionated with gel filtrations and activated thiol Sepharose 4B. The final preparation with D activity was proved to correspond with Band 3 which was not stained with PAS reagent. Deoxycholic acid solubilized stroma was fractionated with gel filtrations and affinity chromatography, resulting in the preparation corresponding to Band 3 with potent Rh-Hr blood type activities. The activities were detected without reductants and the preparation from D negative red cells had no D activity. Rh-Hr blood type activities other than D were observed in the same fraction.

    RIKIISHI, H. and KUMAGAI, K. Sequential Mutagenesis of Drug Resistance in Streptococcus mutans during Synchronous Replication. Tohoku J. exp. Med., 1983, 141 (2), 155-161-The frequency of nitrosoguanidine-induced mutation for streptomycin-, bacitracin- and rifampicin-resistance was measured in Streptococcus mutans during synchronous replication after release from chloramphenicol inhibition. A clear peak of mutagenesis for each marker was observed at certain times during synchronous replication. These times were different for individual markers; the times of peaks for streptomycin-, bacitracin- and rifampicin-resist-ance were 13, 22 and 12min, respectively. At a definite time after the first peak, there was a second one. The distances between the first and second peaks during the synchronous replication were identical for all markers and approximately 5omin which represents the doubling time of the organisms. These results indicate that nitrosoguanidine causes sequential mutagenesis for these three markers in Streptococcus mutans when the growth is resumed after chromosome alignment, so that the methods may be useful in determining the sequence of gene replication for various markers in Streptococcus mutans.

Med., 1983, 141 (2), 163-173-The immunofluorescence technique using antisera against human plantar stratum corneum fibrous protein (total keratin) and 64K M.W. keratin subunit isolated from total keratin by SDS polyacrylamide gel electrophoresis (PAGE) was used to examine epidermal keratinization in some skin disorders. In normal skin, total keratin was distributed throughout the epidermis, while 64K keratin was localized at the suprabasal layers. In the case of squamous cell carcinoma (SCC), the tumor cells were uniformly stained positive with total keratin antiserum, whereas diminished or negatively stained cells were observed with 64K keratin antiserum (64K antiserum), suggesting that the tumor included cells in various stages of differentiation. In basal cell epithelioma (BCE), most of the tumor cells were negatively stained with 64K antiserum being consistent with the histologic observation that BCE is originated from the basal cells. However, some of the tumor cells were stained positive with 64K antiserum, indicating that individual cell keratinization might occur in BCE. In lichen planus, an inflammatory disease demonstrating so-called lichnoid tissue reaction, positively stained colloid bodies in the upper dermis were observed either with total keratin antiserum or with 64K antiserum. It was suggested that colloid bodies resulted from individual keratinization of damaged keratinocytes during inflammation.

Effects o f Radiotherapy and Chemotherapy on Sister Chromatid Exchanges in Peripheral Lymphocytes of the Patients with Gynecologic Cancer. Tohoku J. exp. Med., 1983, 141 (2), 175-181-Studies were done on the influence of radiotherapy or chemotherapy on peripheral lymphocyte SCE (sister chromatid exchange) frequency in patients with gynecologic cancer. The average value of spontaneous SCE frequency in 10 patients who had received radiotherapy was 6.94±2.57 (mean±S.D.), which was significantly lower than the control value of 8.59±2.99 (p<0.001). The rate of decline of these frequency values was negatively correlated with the irradiation dosage. In contrast, the average spontaneous SCE frequency in the patients who had received combination chemotherapy was 12.48±4.02, which was significantly higher than the control value (p<0.001). The average value of MMC-induced SCE frequency in patients under chemotherapy was much the same as the control value of 26.16±5.77. That in patients under radiotherapy was 23.88±5.65, which was slightly lower than the control value. There was, however, no significant difference in the MMC-induced SCE frequency value among these 3 groups.

Med., 1983, 141 (2), 183-190-Several enzyme activities involved in lipid hydrolysis (acid and neutral cholesterol esterase and lipase) or synthesis (aryl-CoA synthetase, acyl-CoA: cholesterol acyltransferase and cholinephosphotransferase) were assayed in the aortas of rats with nephrosis induced by daunomycin and of rats fed on high cholesterol diet. In nephrotic rats activities of some enzymes involved in lipid hydrolysis, but not in synthesis, were increased. On the contrary, in rats fed on high cholesterol diet, the activities of all enzymes involved in lipid synthesis were significantly increased, with some increase in those involved in lipid hydrolysis. In nephrotic rats fed on high cholesterol diet all enzyme activities were markedly increased. From the view point of accumulation of cholesterol ester (CE), the ratio of hydrolysis of CE in lysosomes to CE incorporated from the blood and the ratio of hydrolysis of CE to reesterification of free cholesterol in microsomes were considered to be important. From this point of view, nephrotic hyperlipidemia was not so atherogenic as hyperlipidemia induced by the diet. The role of serum high density lipoproteins in lipid metabolism in the aorta was discussed.

Med., 1983, 141 (2), 191-197-The lipid contents of the plasma and liver and enzyme activities in the aorta were determined in rats given the following diets: Group I, normal diet (ND); Group II, high cholesterol diet (HCD, ND plus 1% cholesterol and 0.5% cholic acid); Group III, HCD plus 0.5% γ-oryzanol; Group IV, HCD plus 2% γ-oryzanol in the last days of the 4th, 8th and 12th week of the feeding period. Total cholesterol, low-density lipoprotein-cholesterol and very low-density lipoprotein-cholesterol in the plasma were significantly higher in rats in groups III and IV than in those in group II. High-density lipoprotein-cholesterol was increased in groups III and IV. There were no differences in the triglyceride or phospholipids concentrations in the plasma of rats in the four groups. The liver weight was higher in the HCD groups (groups II, III and IV) than in the ND group (group I). The cholesterol ester and triglyceride contents of the liver were higher in the HCD groups than in the ND group, and the increase was the least in group IV of the HCD groups. Changes in acid choleterol esterase activity and acyl CoA: cholesterol acyltransferase activity in rat aorta homogenates to compensate for lipid deposition were greater in groups III and IV than in group II.

Tohoku J. exp. Med., 1983, 141 (2), 199-205-The important function of Sialic acids in glycoproteins is to prolong the half life of the glycoproteins in the circulation. In this study, the sialic acid content of plasma low density lipoprotein (LDL)-apoproteins (predominantly B-100 apoprotein, a sialoglycoprotein) was analyzed together with relative electrophoretic mobility of LDL in 46 hypercholesterolemic and normocholesterolemic diabetics and non-diabetics. There was a significant positive correlation between the LDL-sialic acid content and LDL mobility either in the non-diabetics (r=0.534, p<0.01) or in the diabetics (r=0.482, p<0.02). Thus, the effect of the LDL-sialic acids upon the LDL electric charge was apparent. However, the LDL-sialic acids in the hypercholesterolemic diabetics were significantly (p<0.01) decreased to 9.64±0.63μg/mg LDL protein (mean±S.E.) as compared with that in the normocholesterolemic diabetics (12.85±0.62μg/mg LDL protein). Since the relatively sialic acid-poor LDL particles are rather accumulated in plasma of diabetic patients, the grade of sialylation of glycoprotein seems to have no role in the interaction with peripheral cells and therefore, the glycoprotein turnover, as far as the LDL apoproteins are concerned.

Simultaneous Measurement of Human Prolactin and Growth Hormone Levels in Maternal and Fetal Serum and Amniotic Fluid. Tohoku J. exp. Med., 1983, 141 (2), 207-210-Growth hormone (GH) and prolactin (PRL) in maternal and fetal serum and amniotic fluid were measured simultaneously by radioimmunoassay at term deliveries of 20 normal mothers. The PRL level of amniotic fluid was significantly higher than that of maternal and fetal serum. The fetal GH level was significantly higher than the maternal and amniotic levels of GH. There was no significant correlation among hormone levels of maternal and fetal sera and amniotic fluid. These data suggest that PRL and GH levels of maternal and fetal sera and amniotic fluid are controlled separately.

Med., 1983, 141 (2), 211-215-Rabbit antisera were prepared by immunization with O red cells or O.Se saliva. Anti-O red cells (anti-Hr) were diluted with O.Se saliva and each of diluents was adopted for elution method on blood or body fluid stains. Anti-Hr gave positive reactions only with red cell stains. On the other hand, anti-O.Se (anti-Hs) absorbed with O red cells kept activity reacting with latex coated with O.Se saliva. Anti-Hs reacted with various body fluid stains but not with red cell stain. It is considered that anti-Hr and -Hs are useful tools for individualization of blood or body fluid stains in forensic practice.

Med., 1983, 141 (2), 217-224-Stroma from D positive red cell was solubilized with 1% deoxycholic acid (DOC) and was applied onto Sepharose 4B and 6B, followed by fractionation with activated thiol Sepharose 4B. By this method Band 3 containing no carbohydrate was prepared. The Band 3 was incubated with human anti-D, then immune complex soluble in DOC solution was produced. This complex was not prepared from red cell components other than Band 3, nor from Band 3 from D negative red cell. The complex agglutinated papainized D positive red cells. Molecular weight of the complex was estimated to be 500, 000 to 700, 000 by the gel filtration method. The molecular weight increased according to the time of incubation. The complex gave a precipitation line in counter immunoelectrophoresis (IEP), a single peak in crossed IEP against anti-Band 3, and a line at γ-globulin region in IEP against anti-human serum. SDS-polyacrylamide gel disc electrophoresis of the complex showed the pattern consisting of fragments of immunoglobulin G and Band 3 which was not stained with periodic acid Schiff reagent. These support our results that Rh-Hr blood type activities are located in Band 3 almost free of carbohydrate.

Med., 1983, 141 (2), 225-235-There are two methods for hemolytic titration of C4. One is a method using EAClgp oxyC2hu and EDTA-Cgp (E-C method) and the other using EAClgp, oxyC2hu and C4Dgp (C4D mehtod). The C4 titers obtained by these two methods differed depending on whether the C4 exists in plasma or in serum. In the case of Ssh strain, serum C4 activity assayed by the E-C method was higher than that by the C4D method, while plasma C4 activity assayed by the E-C method was lower than that by the C4D method. It was hypothesized that these results would occur from the difference in the state of C4 molecule present in serum or plasma. From chromatographic analysis, it was apparent that the C4 molecule in serum existed in at least four different molecular states; free C4 molecule with low Ss antigenicity, hemolytically inactive Ss protein, active C4 with high molecular weight and low Ss antigenicity, and inactive Ss protein with high molecular weight. Their expression of Ss antigen and the hemolytic C4 activity were different, and each type of C4 molecule behaved differently in the assay of the two methods. Furthermore an important role of mouse C5 in the hemolytic assay of mouse C4 is discussed.

Med., 1983, 141 (2), 237-239-A modified mixed agglutination method is described for detecting AB antigen from body fluid stain. Saline extract of body fluid stain was added to A.Se saliva stain which was beforehand sensitized with anti-A. AB antigen reacting with the stain was detected with the usual mixed agglutination method using anti-B and B red cells.

Med., 1983, 141(2), 241-242-Influence of contralateral finger motions on ipsilateral finger extension was analyzed in order to explore optimal condition of bilateral motions as a facilitation technique, measuring motor times (MTs) of finger extensor muscles in four conditions; homonymous or non-homonymous as for muscle coupling and simultaneous or successive as for timing of motion-initiation. Compared to unilateral motion, MTs shortened only in bilateral motion with preceding sustained contractions of the contralateral homonymous muscle.

Tohoku J. exp. Med., 1983, 141 (2), 243-244-The urethral pressure profile (UPP) was recorded in anterior, posterior, and right and left lateral portions of the canine urethra using a newly designed catheter with only one eye around the circumference of the tip for UPP recording. Our single-eye catheter made it possible to measure UPP in the four aspects separately and confirmed that there was a directional difference in UPP.

Med., 1983, 141 (2), 245-246-Conduction velocity of excitation in various regions of the canine pelviureter was studied through the simultaneous recordings of electromyograms at four sites of the canine pelvicalyceal preparation. The pelviureteral system was maintained in the condition which was similar to in vivo situation by infusion of oxygenated Krebs-Ringer solution into the renal pelvis at the average flow rate of living dogs. The conduction velocity was slowest in the proximal region of the pelvis where the pacemaker was located, i.e., about 5.8mm/sec on an average. The velocity gradually increased in the distal region of the pelvis and the ureter. It was suggested that the conduction velocity of excitation was significantly different in the proximal area of the pelvis, the distal area of the pelvis and the ureter.

    IGARASHI, M. The Activity of Arylsulphatases in Serum of Patients with Several Bullous Diseases. Tohoku J. exp. Med., 1983, 141 (2), 247-248-Spectrophotometric estimations of levels of activities of arylsulphatases A and B in serum of patients with several bullous diseases were carried out. The activities of these enzymes in cases of bullous pemphigoid were at high levels in comparison with those in cases of other bullous diseases and in the control group. Especially, the activity of arylsulphatase B was remarkably increased. It seems likely that there is some relationship between these disturbances and the pathogenesis of bullous pemphigoid.
